Summary: This study reports the synthesis and in vitro evaluation of a series of compounds against Leishmania donovani. The most active compound showed promising antileishmanial activity and stability in rat liver microsomes. Molecular modeling studies suggest a possible binding mechanism with Leishmania donovani topoisomerase 1. These findings highlight the potential of these compounds as antileishmanial agents.

Summary: A series of rosmarinic acid-beta-amino-alpha-ketoamide hybrids were synthesized and showed promising activity against Leishmania donovani. Two hybrids, 2g and 2h, exhibited comparable anti-leishmanial activity to erufosine but had no cytotoxic effects on human THP-1 cells. Molecular docking studies emphasized the importance of the aliphatic chain at the alpha-carbon of the cinnamoyl carbonyl group for activity.

Summary: In this study, an inclusion complex (IC6HKTZ) of ketoconazole (KTZ) with p-sulfonic acid calix[6]arene (CX6SO3H) was prepared to improve the solubility and efficacy of KTZ against Leishmania amazonensis and Leishmania infantum promastigotes. The solubility enhancement of KTZ by 120.00 mu M CX6SO3H was about 95 times. IC6HKTZ exhibited lower IC50 values compared to free KTZ for both Leishmania species, indicating improved anti-leishmanial activities.

Summary: Leishmaniasis is a neglected tropical disease with limited treatment options. The discovery of 2,4,5-trisubstituted benzamides as potent antileishmanial compounds with low aqueous solubility has opened up new possibilities for drug development. Through extensive studies on structure-activity and structure-property relationships, early leads with suitable potency, stability, and solubility were identified. One of the early leads, compound 79, showed high oral bioavailability and effectively inhibited Leishmania growth in animal models. These benzamide compounds hold promising potential for the development of orally available antileishmanial drugs.
