Summary: Isolation and genetic characterization of Toxoplasma gondii strains from domestic animals in Southern and coastal area of Tunisia revealed a large predominance of type II strains, followed by type III, and for the first time in Tunisia, an isolate of Africa 4 lineage. Analyses of population genetics also showed the presence of a divergent population of type II lineage in Tunisia, supporting limited recent migrations of strains between Tunisia and other countries.

Summary: The study found that TLR2(-/-) mice are less susceptible to abnormal pregnancy induced by Toxoplasma gondii infection compared to wild-type mice. While there were no significant differences in parasite numbers and histological changes between the two groups, the expression of certain cytokines in the placentas of infected wild-type mice was affected by T. gondii infection, while TLR2(-/-) mice showed no such changes. Serum interferon-gamma levels were significantly lower in infected TLR2(-/-) mice, indicating a potential role of TLR2 signaling in immune response and placental function during late pregnancy.

Summary: Discovery of the protein BCLA/MAG2 as a specific marker for bradyzoite and cyst envelope in T. gondii has been achieved through genetically engineered parasites. By using lab mice as models, researchers have confirmed that BCLA/MAG2 can stimulate antibody production recognizing bradyzoite and cyst structure. An ELISA assay based on a bacterially produced BCLA recombinant polypeptide has been designed and validated using sera from mice and human patients with toxoplasmosis, showing promising results for detection of persistent T. gondii infection.

Summary: In this study, the impact of T. cruzi and recombinant P21 on T. gondii infection was investigated in BeWo cells and human placental explants. The results showed that T. cruzi infection and rP21 can increase the invasion and decrease the intracellular proliferation of T. gondii in BeWo cells. In human placental villi, rP21 can either increase or decrease T. gondii proliferation, while T. cruzi infection increases T. gondii proliferation. These findings highlight the importance of neonatal screening for both diseases and suggest P21 as a potential therapeutic target for Chagas disease.
