Summary: This study compared the foot kinematics of toe walking and heel-toe walking in healthy individuals, finding increases in forefoot plantarflexion and hindfoot internal rotation during toe walking compared to heel-toe walking. These results may aid in distinguishing the pathologic mechanism of equinus gait in various disorders.

Summary: ENPP1 is frequently overexpressed in local relapses and tumor metastases, promoting an immunosuppressive tumor micro-environment and impairing immune response. Blocking ENPP1 could enhance immune remodeling and activate the STING pathway. Combination therapy with ENPP1 inhibitors and other modalities shows potential in improving antitumor-immune responses and clinical outcomes. This review summarizes the current state of the art and provides new perspectives for novel treatment strategies.

Summary: In this study, VH single-domain antibodies against ENPP1 were generated using phage and yeast display. A VH domain was discovered that allosterically inhibited the hydrolysis of cGAMP and ATP. Furthermore, the VH domain was engineered into multispecific formats and immunotherapies, showing potent cellular activity.

Summary: ENPP1, a type II transmembrane glycoprotein, is highly expressed in various cancers and associated with poor prognosis. In this study, two high-affinity and specific anti-ENPP1 Fab antibodies, 17 and 3G12, were isolated and characterized. Antibody-drug conjugates, IgG-based bispecific T-cell engagers, and CAR T-cells were generated based on these antibodies, all demonstrating potent killing of ENPP1-expressing cells. These antibody-derived modalities hold promise as therapeutic candidates for cancers expressing human ENPP1.
