Colorectal cancer (CRC) is a serious public health problem that results due to changes of diet and various environmental stress factors in the world. Curcumin is a traditional medicine used for treatment of a wide variety of tumors. However, antimetastasis mechanism of curcumin on CRC has not yet been completely investigated. Here, we explored the underlying molecular mechanisms of curcumin on metastasis of CRC cells in vitro and in vivo. Curcumin significantly inhibits cell migration, invasion, and colony formation in vitro and reduces tumor growth and liver metastasis in vivo. We found that curcumin suppresses Sp-1 transcriptional activity and Sp-1 regulated genes including ADEM10, calmodulin, EPHB2, HDAC4, and SEPP1 in CRC cells. Curcumin inhibits focal adhesion kinase (FAK) phosphorylation and enhances the expressions of several extracellular matrix components which play a critical role in invasion and metastasis. Curcumin reduces CD24 expression in a dose-dependentmanner in CRC cells. Moreover, E-cadherin expression is upregulated by curcumin and serves as an inhibitor of EMT. These results suggest that curcumin executes its antimetastasis function through downregulation of Sp-1, FAK, and CD24 and by promoting E-cadherin expression in CRC cells.

PCDH10 exerts tumor-suppressor functions through modulation of EGFR/AKT axis in colorectal cancer

Tzu-Ming Jao, Woei-Horng Fang, Shih-Ci Ciou, Sung-Liang Yu, Yu-Lin Hung, Wei-Ting Weng, Tsai-Yi Lin, Ming-Hong Tsai, Ya-Chien Yang

Summary: PCDH10 is identified as a tumor suppressor that induces cell cycle retardation and apoptosis in colorectal cancer cells, reverses epithelial-mesenchymal transition process, and inhibits stemness of CRC cells. PCDH10 inhibits AKT/GSK3 beta signaling pathway, reduces beta-catenin activity, and directly engages in negative regulation of EGFR/AKT/beta-catenin signaling pathway for tumor suppression.

Summary: This study assessed the performance of PD-L1 diagnostic assays in NSCLC, HNSCC, and UC. The results showed that the performance of Ventana SP263 and Dako 22C3 assays was comparable in NSCLC and HNSCC. However, in UC, the performance of Ventana SP263 assay was less reliable and may impact treatment decisions.

???????Regulation of dendritic cell maturation in osimertinib-treated lung adenocarcinoma patients

Ming-Fang Wu, Ya-Hsuan Chang, Hsuan-Yu Chen, Chao-Chi Ho, Huei-Wen Chen

Summary: Osimertinib is effective for lung adenocarcinoma patients with EGFR mutations, but combining it with immune checkpoint inhibitors increases the risk of interstitial lung disease. This study investigates the interaction between dendritic cells and osimertinib and suggests that osimertinib therapy may activate dendritic cell functions, potentially leading to increased immune toxicity when combined with immunotherapy.

LCRMP-1 is required for spermatogenesis and stabilises spermatid F-actin organization via the PI3K-Akt pathway

Jung-Hsuan Chang, Chia-Hua Chou, Jui-Ching Wu, Keng-Mao Liao, Wei-Jia Luo, Wei-Lun Hsu, Xuan-Ren Chen, Sung-Liang Yu, Szu-Hua Pan, Pan-Chyr Yang, Kang-Yi Su

Summary: Mice deficient in LCRMP-1 show reduced fertility and perturbed F-actin cytoskeleton dynamics via the PI3K-Akt-mTOR axis. LCRMP-1 is abundantly expressed in the testis and plays a crucial role in spermatogenesis. LCRMP-1 deficiency causes disorganized F-actin due to unbalanced signaling of F-actin dynamics through upregulated PI3K-Akt-mTOR signaling, leading to aberrant spermiation and reduced fertility.
