Due to their properties and characteristics human mesenchymal stem cells (MSCs) appear to have great therapeutic potential. Many different populations of MSCs have been described and to understand whether they have equivalent biological properties is a critical issue for their therapeutic application. We proposed to analyze the in vitro growth kinetics of MSCs derived from different body sites (iliac crest bone marrow, vertebrae bone marrow, colon mucosa, dental pulp). Mesenchymal stem cells derived from vertebrae can be maintained in culture for a greater number of steps and they also generate mature cells of all mesenchymal lineages with greater efficiency, when induced into osteogenic, adipogenic and chondrogenic differentiation. The ability of vertebrae-derived MSCs in terms of expansion and differentiation is very interesting at the light of a clinical application for bone fusion in spine surgery.

Giovanni Barbanti Brodano, Cristiana Griffoni, Giancarlo Facchini, Elisa Carretta, Francesca Salamanna, Rohan Gala, Giuseppe Tedesco, Gisberto Evangelisti, Silvia Terzi, Riccardo Ghermandi, Stefano Bandiera, Marco Girolami, Valerio Pipola, Milena Fini, Alessandro Gasbarrini

Summary: The retrospective analysis of 14 patients treated with bone marrow concentrated (BMC) injection for aneurysmal bone cyst (ABC) of the spine showed that BMC injection can promote ossification of the cyst and improve clinical outcomes. The study emphasized the importance of BMC as a valid alternative for spinal ABC management.

Luca Gianaroli, Amerigo Vitagliano, Anna P. Ferraretti, Silvia Azzena, Gaia Terzuoli, Davide Perruzza, Guido Ambrosini, Carla Tabanelli, M. Cristina Magli

Summary: This study investigated the effectiveness and safety of a mild ovarian stimulation program called IVF Lite in good prognosis patients. The results showed that the program was effective and safe for patients with a good response to clomiphene citrate stimulation, and two-thirds of the patients achieved a live birth at the completion of the program.

Summary: This study aimed to translate and validate the Italian version of the Spine Oncology Study Group Outcome Questionnaire (SOSGOQ), a specific tool for measuring health-related quality of life in Italian-speaking patients with spinal metastases. The results of the study showed that the Italian version of the SOSGOQ had high reliability and validity among Italian-speaking patients affected by spinal metastases.

Birefringence properties of human immotile spermatozoa and ICSI outcome

M. Cristina Magli, Andor Crippa, Davide Perruzza, Silvia Azzena, Serena Graziosi, Francesco Coppola, Carla Tabanelli, Anna P. Ferraretti, Luca Gianaroli

Summary: Selecting sperm cells with birefringence properties under polarized light is a successful strategy to identify viable spermatozoa. This approach improves fertilization, embryo development, and implantation rates compared to non-birefringent cells, leading to higher cumulative live birth rates.

Failure to detect DNA in blastocoel fluid is associated with a higher live birth rate in both PGT-A and conventional IVF/ICSI cycles

L. Gianaroli, D. Perruzza, C. Albanese, S. Azzena, C. Tabanelli, Anna P. Ferraretti, M. Cristina Magli

Summary: This study found that the presence of DNA in the blastocoel fluid (BF) of expanded blastocysts, assessed by whole genome amplification (WGA), is associated with the clinical outcome at the first transfer. Blastocysts with negative BF-WGA have a higher chance of implanting and developing to term compared to those with positive BF-WGA, in both preimplantation genetic testing for aneuploidies (PGT-A) cycles and in IVF/ICSI conventional cycles.

Summary: This retrospective study aimed to evaluate the overall survival of surgically treated patients with lung cancer spinal metastases and identify factors associated with better survival. The results showed that surgical treatment can improve the quality of life and control pain, thereby enhancing autonomy.

Summary: The study found that known chromosome segregation errors accounted for 90.4% of abnormal chromosome copy numbers in polar bodies of oocytes from women of advanced maternal age. These errors were associated with impaired embryo development.

Gender-Specific Differences in Human Vertebral Bone Marrow Clot

Francesca Salamanna, Deyanira Contartese, Veronica Borsari, Stefania Pagani, Maria Sartori, Matilde Tschon, Cristiana Griffoni, Gianluca Giavaresi, Giuseppe Tedesco, Giovanni Barbanti Brodano, Alessandro Gasbarrini, Milena Fini

Summary: Recently, our group studied the use of vertebral bone marrow (vBMA) clot as a cell therapy for spinal fusion. We compared the properties of vBMA clots and derived vertebral mesenchymal stem cells (MSCs) from female and male patients who underwent spinal fusion procedures and received vBMA clot treatment. The results showed that vBMA clots and MSCs from males had higher expression of growth factors (GFs) and greater osteogenic and chondrogenic potential compared to females. The findings also revealed differences in the gene expression profiles of MSCs derived from vBMA clots between female and male donors.

Summary: CFR-PEEK is increasingly popular in spinal oncological applications due to its advantages over titanium. In this study, CFR-PEEK instrumentation was used to treat lumbar degenerative diseases (DDs). The results showed high fusion rates and significant improvement in clinical outcomes. Carbon fiber implants were well-tolerated and had no mechanical complications. This pilot study supports the efficacy and safety of CFR-PEEK implants in spinal degenerative diseases.

EuMAR: a roadmap towards a prospective, cycle-by-cycle registry of medically assisted reproduction in Europe†

Summary: More than 20 years ago, the European IVF Monitoring initiative began surveying activities in medically assisted reproduction (MAR) and issuing annual reports. These reports have contributed to the transparency and surveillance of reproductive care. Changes in treatment modalities and the introduction of new technologies necessitate a shift towards cumulative outcome results and a data registry for MAR activities. The EuMAR project aims to establish a transnational registry for prospective cycle-by-cycle MAR and fertility preservation data, funded by the European Union.
