Inhibition of Rho kinase by GSK 269962 reverses both corticosterone-induced detrusor overactivity and depression-like behaviour in rats

Literature data give clear evidence that upregulated RhoA/Rho-kinase signalling is one of the factors that may lead to the development of detrusor overactivity and various disorders of the central nervous system. Therefore, the main objective of our study was to investigate whether administration of a Rho-kinase inhibitor - GSK 269962 could reverse corticosterone-induced depressive-like behaviour and changes in cystometric parameters associated with detrusor overactivity, as well as undo the alterations of several biomarkers related to both disorders (i.e., pro-inflammatory/anti-inflammatory cytokines and neurotrophins) in serum, urinary bladder, and different brain structures. The experiments were carried out on female Wistar rats. Surgical procedures, cystometric investigations, biochemical analyses, and behavioural studies (measurement of the locomotor activity and the forced swim test) were performed according to the published literature. As expected, administration of corticosterone at a daily dose of 20 mg/kg for 14 days increased the immobility time of animals in the forced swim test, induced changes in the cystometric parameters specific to bladder overactivity, reduced levels of neurotrophins, and elevated concentrations of the pro-inflammatory cytokines. Inhibition of Rho-kinase by 7-day treatment with GSK 269962 (10 mg/kg/day) reversed the symptoms of both detrusor overactivity and depression as well as normalized levels of the tested biomarkes. Our findings encourage the idea of Rho-kinase inhibitors as a potential future treatment option for overactive bladder accompanied by depression.

Femistina versus Canephron as a prevention of urinary tract infections after midurethral sling surgery - Non-inferiority study

Evaluation of developmental toxicity in zebrafish embryos and antiproliferative potential against human tumor cell lines of new derivatives containing 4-nitrophenyl group

Summary: The aim of the studies was to evaluate the antiproliferative potential of new derivatives containing 4-nitrophenyl group against human tumor cell lines and its impact on developmental toxicity in zebrafish model. The results showed that APS-1 and APS-18 compounds reduced the viability of human tumor lines, with APS-18 showing strong and selective antiproliferative activity against all neoplastic cell lines, particularly glioblastoma. The fish embryo acute toxicity test demonstrated that APS-1 and APS-18 did not cause morphological abnormalities in zebrafish embryos, indicating their potential for further research and development of safer drugs with better anticancer properties.

Postoperative Prevention of Urinary Tract Infections in Patients after Urogynecological Surgeries-Nonantibiotic Herbal (Canephron) versus Antibiotic Prophylaxis (Fosfomycin Trometamol): A Parallel-Group, Randomized, Noninferiority Experimental Trial

Summary: Urinary tract infections (UTIs) are common complications of urogynecological surgeries, and current preventive methods using antibiotics are becoming less effective due to increasing antibiotic resistance. This study aimed to compare the effectiveness of Canephron, a phytotherapeutic drug, with antibiotic prophylaxis in preventing postoperative UTIs. The results showed that Canephron was noninferior to antibiotic prophylaxis in preventing UTIs, suggesting its potential as an alternative method to decrease antibiotic consumption and combat antibiotic resistance.

The Cannabinoid Ligand Arachidonyl-2′-Chloroethylamide (ACEA) Ameliorates Depressive and Overactive Bladder Symptoms in a Corticosterone-Induced Female Wistar Rat Model

Summary: This paper aimed to investigate whether the selective cannabinoid CB1 receptor agonist ACEA could reverse the effects of corticosterone (CORT) on depressive and overactive bladder potential. Through experiments on 48 female rats, it was found that ACEA could restore CORT-induced bladder dysfunction and depressive behavior, as well as regulate urine and other biomarkers.

Effects of topical dehydroepiandrosterone therapy in women after pelvic organ prolapse surgery

Summary: The objective of this study was to develop intravaginal vaginal suppositories containing DHEA and compare the clinical effects of vaginal topical therapy with DHEA, estradiol, or antibiotic after POP surgery. It was found that both DHEA and estradiol topical therapy improved maturation index, vaginal pH, and vaginal symptoms. Topical therapy with DHEA after pelvic organ prolapse repair brings similar results as estradiol, without potential systemic exposure to increased concentrations of sex steroids above levels observed in postmenopausal women.

Synchronously occurring endometrioid carcinomas of the uterine corpus and uterine cervix preceded by different precancerous lesions: A case study and a literature review

Summary: The occurrence of synchronous carcinomas originating from the uterine corpus and uterine cervix, of the same histological subtype, is extremely rare. This case study presents a synchronous early stage G1 adenocarcinoma of the uterine corpus with cervical G2 endometrioid adenocarcinoma. The tumors differ in histological grading and clinical stage, and are preceded by different precancerous lesions, atypical endometrial hyperplasia and foci of endometriosis localized within the uterine cervix.

Endometrial cancer diagnostic and prognostic algorithms based on proteomics, metabolomics, and clinical data: a systematic review

Summary: Endometrial cancer, the most common gynaecological malignancy, has a high incidence and mortality rate in developed countries. There is a need for non/minimally invasive tools for diagnosis and prognosis to manage the increasing number of women at risk. Biomarkers from proteomics and metabolomics can be used to develop such tools, and this review explores the current research in this field. A systematic review of published studies using proteomics and/or metabolomics for biomarker discovery in endometrial cancer is provided, along with recommendations for future studies.

Evaluation of Antidepressive-like Behaviours and Oxidative Stress Parameters in Mice Receiving Imipramine-Zinc Complex Compound

Summary: The study evaluated the antidepressant-like effects of an IMI-Zn complex compound on mice and compared its efficacy with separate administration of imipramine and zinc. The results showed that the IMI-Zn complex exhibited dose-dependent antidepressant potential similar to individual treatments of imipramine and zinc. The study also found that acute stress did not significantly affect oxidative stress parameters. Developing the IMI-Zn complex could simplify treatment and improve patient compliance.

Laparoscopic Radical Trachelectomy after Neoadjuvant Chemotherapy for Fertility Preservation in Early-Stage Bulky Cervical Cancer: A Case Report and Literature Review

Summary: The management of early-stage cervical cancer in young women poses challenges in preserving fertility and achieving optimal oncological outcomes. Insufficient data hinder proper counseling for this vulnerable patient group. This report presents a case of successful treatment outcome in a young woman with early-stage cervical cancer who desired fertility preservation, using neoadjuvant chemotherapy followed by laparoscopic radical trachelectomy. Ongoing prospective trials are expected to provide stronger evidence in this area.
