Summary: Flow cytometry was used to detect the presence of PMN-MDSCs in the spleen and joints of CIA mice, and their role in the pathogenesis of RA was confirmed through experiments involving PMN-MDSC depletion and adoptive transfer. Our study found that PMN-MDSCs in CIA mice had high expression levels of genes including BAFF, and BAFF promoted TNF-a expression through the BTK/NF-?B signaling pathway. This study reveals a novel pathogenesis of PMN-MDSCs in CIA.

Summary: This study found that curcumin can improve rheumatoid arthritis by inhibiting cell proliferation, migration, invasion, and promoting cell apoptosis. Curcumin also reduces inflammation by decreasing the secretion of inflammatory factors. Therefore, curcumin may be a promising candidate for treating RA.

Summary: The study demonstrated the analgesic and anti-inflammatory effects of HCRG21, a peptide blocker of TRPV1, in a mice model of carrageenan-induced paw edema. HCRG21 effectively inhibited edema formation, reduced TNF-alpha production, and showed potent analgesic effects on mechanical and thermal hyperalgesia models. These results highlight the potential of TRPV1 blockers like HCRG21 in managing pain and inflammation.

Summary: This review discusses the effects of capsaicin on vascular function, indicating that excessive consumption of capsaicin may lead to vasoconstriction and myocardial infarction, while emphasizing the role of inflammation in sensitizing the vascular channels to capsaicin activation.

Summary: Capsaicin, the pungent ingredient in chili peppers, can cause intense burning pain in humans, but it has also been used as an analgesic for chronic pain conditions. The analgesic effects of capsaicin depend on concentration, with low concentrations inducing short-term defunctionalization and high concentrations leading to long-term defunctionalization.

Summary: Autophagy plays a crucial role in regulating the ageing process and neurodegeneration. Dysfunction of autophagy is implicated in Alzheimer's disease pathogenesis. The activation of TRPV1 using capsaicin induces autophagy, which could potentially be a therapeutic target for Alzheimer's disease.

Summary: Microglia energy metabolism is suppressed during chronic A beta-tolerant processes, but pharmacological activation of TRPV1 can rescue microglial dysfunction and restore immune responses, leading to reduced amyloid pathology and reversed memory deficits in AD mice. This study suggests that TRPV1 is an important target for regulating metabolic reprogramming in microglial functions for AD treatment.

Summary: Nitrogen mustard causes severe vesicating skin injury, partially through over-activating autophagy, and partly by activating the TRPV1-Ca2+-CaMKK beta-AMPK-ULK1 signaling pathway. These results suggest that blocking TRPV1-dependent autophagy could be a potential treatment strategy for nitrogen mustard-caused cutaneous injury.

Summary: Capsaicin exhibits significant hypoglycemic effects by activating the TRPV1 channel, which is lost when the channel is blocked. The hypoglycemic effect of capsaicin is partially attributed to the combined up-regulation of TRPV1 receptors and PDX-1.

Summary: Dexmedetomidine (Dex), an analgesic and sedative drug with anti-inflammatory properties, was found to reduce the migration of fibroblast-like synoviocytes (FLSs) in rheumatoid arthritis (RA) and decrease the expression of RA-related factors. This suggests that Dex may be a potential therapeutic drug for RA.

Summary: This study showed that activation of TRPV1 by capsaicin increases the expression and function of NKCC1 protein in DRG, possibly mediated by the PKC/p-ERK signaling pathway. These findings suggest that DRG NKCC1 may participate in the inflammatory pain induced by TRPV1.

Summary: KUS121 is a novel medication developed to protect chondrocytes from stress-induced cell death. It has shown significant effects in both in vitro and in vivo studies, reducing apoptosis and improving gene expression in chondrocytes under stress conditions. These results suggest that KUS121 may be a promising therapeutic agent for preventing the progression of osteoarthritis.
