Neutral Mobius Aromatics: Derivatives of the Pyrrole Congener Aza[11]annulene as Promising Synthetic Targets

Amino- and methyl-disubstituted derivatives of an aromatic 12 pi electron pyrrole homologue Mobius 1H-aza[11]annulene are predicted computationally at the B3LYP/6-31G* level of theory to be distinctly aromatic and a neutral ground state on its potential surface. The parent annulene has a nonaromatic conformation, which is 6.0 kcal mol(-1) lower in energy at the CCSD(T)/D95*//B3LYP/6-31G* level of theory than a structurally very similar Mobius conformation. After derivatization, however, geometry optimizations of the two lowest-energy - Mobius and non-Mobius - heteroannulene conformations both converged two a single Mobius minimum, whereas two (Mobius and non-Mobius) distinct difluoro-substituted species could be located 1.8 kcal mol(-1) apart in energy at the B3LYP level. The Mobius minimum is 7.5 kcal mol(-1) lower in energy at the coupled cluster level than a Huckel topology form derived from a known bridged 1H-[4,9]methano[11]annulene. Several possible reactive modes are investigated in order to assess the relative stability of the Mobius parent annulene. In addition, we report formal novel. examples for charged and neutral Mobius triplet aromatic annulenic and heteroannulenic systems, some of which also exhibit distinct aromatic properties. Further fundamental insights into the phenomenon of Mobius aromaticity are suggested. The aromaticity of Mobius annulenes is for the first: time linked to wavefunction boundary conditions. Summary: In this study, new artesunic acid and synthetic peroxide-based hybrids were synthesized and analyzed for their inhibitory activity against SARS-CoV-2 and leukemia cell lines. The results showed that several artesunic acid-derived hybrids exhibited potent inhibition against both K562 leukemia cells and SARS-CoV-2. These findings provide valuable insights for the design of artemisinin-derived hybrids to treat cancer and SARS-CoV-2 infections.

Chemical hybridization of sulfasalazine and dihydroartemisinin promotes brain tumor cell death

Lewis Acids and Heteropoly Acids in the Synthesis of Organic Peroxides

Summary: This article summarizes the main achievements in the synthesis of organic peroxides using Lewis acids and heteropoly acids for the first time. It is divided into three parts: (1) the application of metal-based Lewis acids in the synthesis of organic peroxides; (2) the synthesis of organic peroxides promoted by non-metal-based Lewis acids; and (3) the application of heteropoly acids in the synthesis of organic peroxides. The information covered in this review will be useful for specialists in organic synthesis, reactions and processes of oxygen-containing compounds, catalysis, pharmaceuticals, and materials engineering.

Visible-Light-Driven Metal-Free C-H Functionalization: Access to New Bioactive Tetrahydroisoquinoline-Butenolide Hybrids via Domino Amine Oxidation/Vinylogous Mannich Reaction

Summary: An efficient metal-free visible-light-driven two-step domino reaction method was developed for the synthesis of new bioactive compounds. The combination of fluorescein and thiourea was found to be the most effective way to promote the reaction, achieving high yields with a broad substrate scope. Experimental and computational evidence supported the crucial role of singlet oxygen in the reaction, while thiourea acted as an electron-transfer mediator and/or scavenger of reactive oxygen species. Furthermore, the synthesized compounds showed high antischistosomal and anti-cancer activities.

The approved antiviral drugs remdesivir, molnupiravir, and Paxlovid have broad effectiveness, but none are recommended for prophylactic use, highlighting the need for new small molecules and antiviral strategies.
