Summary: NK cells integrate heterogeneous signals for activation and inhibition using germline-encoded receptors, which may be key to comprehensive cancer control. Strategies to leverage their naturally occurring diversity and agility have not yet been developed. Their receptors and signaling pathways play important roles in cancer and potential as targets for immunotherapies.

Summary: As the placenta is semi-allogenic, it is expected that maternal immune cells would attack the invading trophoblast, but studies have shown that they cooperate with the trophoblast to disrupt the arterial wall. uNK cells, unique immune cells, play a role in pregnancy. The interaction between HLA class 1b genes and maternal uNK cells in humans is described in this review.

Summary: Natural Killer (NK) cells have evolved significantly since their initial discovery in the 1970s. Recent research has revealed adaptive-like behavior in NK cells, challenging the traditional categorization of the immune system. This subpopulation of NK cells exhibits characteristics of both innate and adaptive immunity, with clonal-like expansion and enhanced cytotoxic response upon antigen exposure. The implications of these findings are highly relevant in clinical settings, particularly in adoptive immunotherapies where NK cells offer distinct advantages. This review provides an overview of the discovery and current understanding of adaptive NK cells.

Summary: This study investigated the impact of invasive spine surgery on the relationship between monocytes and NK cells in response to Staphylococcus aureus. The findings showed that NK cells continuously lost their ability to produce IFN-gamma during the first week after surgery independently from monocyte-derived IL-12 secretion. Additionally, pre-operative levels of GDF-15 in the serum were associated with the extent of NK cell suppression and hospitalization.

Summary: The study successfully achieved 94% knockout efficiency of CBLB using CRISPR/Cas9 technology in placental CD34(+) cells, demonstrating the negative regulatory role of CBLB in PNK cell cytotoxicity against tumor cells. Ablation of CBLB did not impact PNK cell proliferation, differentiation, or phenotypical characteristics, but resulted in enhanced cytotoxicity against tumor cells. Furthermore, CBLB knockout PNK cells showed increased in vivo proliferation, maturation, and antitumor activity compared to unmodified PNK cells.

Summary: This study aimed to determine whether oridonin enhances the anti-tumor activity of natural killer (NK) cells against lung cancer cells. The results showed that oridonin enhances NK cell cytotoxicity against A549 lung cancer cells by upregulating the expression of degranulation marker CD107a and IFN-γ, as well as activating NK cells and their ligand MICA/B. Oridonin also inhibits STAT3 phosphorylation in A549 cells and NK cells, and has a synergistic effect on the anti-tumor activity of NK cells.

Summary: Millions of Americans infected with COVID-19 require oncologic surgery, and surgery may affect postoperative neuropsychiatric outcomes. This study aimed to investigate whether patients with a history of COVID-19 have an increased risk of developing postoperative delirium after major elective oncologic surgery. The results showed that a preoperative diagnosis of COVID-19 did not increase the risk of postoperative antipsychotic medication use or neurological complications. However, COVID-19 patients had a higher risk of respiratory and overall 30-day complications, indicating a need for further research on post-COVID-19 neurological events.

Summary: The perioperative period has been extensively studied in terms of its impact on oncological outcomes. Basic science research suggests that surgical stress, anesthetics, and analgesics play a role in cancer progression. Retrospective studies have explored the influence of surgery type, anesthetics, analgesics, and blood transfusions on patient survival, but randomized controlled trials have not produced consistent clinical results. This review highlights the challenges of translating basic science into clinical outcomes and identifies future research opportunities.
