Summary: The catalytic properties of small-molecule proteolysis targeting chimeras (PROTACs) may lead to uncontrolled degradation. The main disadvantages of PROTACs are non-cancer specificity and relatively high toxicity, which limit their clinical application. Photocontrolled PROTACs (photoPROTACs) can be triggered by ultraviolet A (UVA) or visible light to induce the degradation of the target protein. A study successfully designed photoPROTACs to degrade bromodomain-containing protein 4 (BRD4) using 365 nm light, with compound N2 proving effective in inducing BRD4 degradation and inhibiting tumor growth in a zebrafish xenograft model of skin cancer tongue squamous cell carcinoma (TSCC) in a photocontrol manner.

Summary: This study investigates the predominance of HSV-1 in oral HSV infections and its effect on oral tongue squamous cell carcinoma (OTSCC). The results show that HSV-1 is the predominant type in oral HSV infections and is also detected in OTSCC samples without clinical significance. Furthermore, low doses of HSV-1 do not affect cell survival or invasion in OTSCC.

Summary: Evidence has shown that 7SK is down-regulated in human tongue squamous carcinoma (TSCC) and acts as a TSCC suppressor through multiple cell-based assays including a migration assay and a xenograft mouse model. This study identified 27 7SK-regulated genes involved in tumor regulation, with upstream regulatory regions bound by 7SK, and suggested potential involvement of transcription factors FOXJ3 and THRA in 7SK-mediated TSCC progression. FOXJ3 and THRA were found to dramatically promote migration in SCC15 cells, potentially contributing to TSCC progression.

Positive margins predict poor recurrence-free survival and local recurrence-free survival. Margins' status was the only independent predictor of local recurrence, while tumor depth of invasion, nodal and margin status were key prognosticators of survival.
