SMAD3 is essential for transforming growth factor-β1-induced urokinase type plasminogen activator expression and migration in transformed keratinocytes

Transforming growth factor-beta 1 (TGF-beta 1) stimulates the extracellular matrix degrading proteases expression and cell migration in order to enhance cancer cells malignancy. In the present study, we analysed the role of TGF-beta 1-induced Smad3 activation in the urokinase type plasminogen activator (uPA) production, as well as in cell migration and E-cadherin downregulation in transformed PDV keratinocyte cell line. TGF-beta 1 signalling was interfered by the chemical inhibitor of the TGF-beta 1-receptor 1 (ALK5), SB505124, and the specific Smad3 inhibitor, SiS3. Our results showed that TGF-beta 1 stimulates uPA expression directly through ALK5 activation. The inhibition of Smad3 strongly reduced the capacity of TGF-beta 1 to stimulate uPA expression, in parallel decreasing the uPA inhibitor plasminogen activator inhibitor type 1 (PAI-1) expression. In addition, the transient expression of dominant negative Smad3 mutant inhibited the TGF-beta 1-induced uPA promoter transactivation. Moreover, Smad3-/- mouse embryonic fibroblasts were refractory to the induction of uPA by TGF-beta 1. The inhibition of both ALK5 and Smad3 dramatically blocked the TGF-beta 1-stimulated E-cadherin downregulation, F-actin reorganisation and migration of PDV cells. Taken together, our results suggest that the TGF-beta 1-induced activation of Smad3 is the critical step for the uPA upregulation and E-cadherin downregulation, which are the key events preceding the induction of cell migration by TGF-beta 1 in transformed cells. Targeting Histone Deacetylases: Opportunities for Cancer Treatment and Chemoprevention

Summary: The dysregulation of gene expression plays a critical role in tumorigenesis, and abnormal activation of histone deacetylases (HDAC) has been reported to cause aberrant histone and non-histone acetylation modifications in hematologic and solid types of cancer. Cancer-associated epigenetic alterations, such as HDAC inhibitors (HDACi), have shown promising potential for anticancer therapy and chemoprevention. This review focuses on the biological roles of the Zinc-dependent HDAC family in malignant diseases, and discusses the use of small molecules and natural products as HDACi for cancer therapy. Structural considerations and clinical trials combining HDACi with current therapies are also discussed, highlighting the potential for future clinical applications of HDACi in precision cancer therapies.

Summary: The tumor suppressor p53 plays a crucial role in controlling liver glycogen storage and is linked to insulin signaling through the transcription factor FOXO1. Acute deletion of p53 prevents glycogen accumulation upon refeeding, while chronic loss of p53 leads to compensational activation of glycogen synthesis pathway. Additionally, high-fat diet-induced insulin resistance with persistent FOXO1 activation affects the regulation of p53 and its target genes during fasting, showing overlapping effects of both FOXO1 and p53 on gene expression in a context-dependent manner.

Summary: Sepsis syndrome is characterized by enhanced secretion of pro-inflammatory cytokines and reactive oxygen species, leading to vascular hyperpermeability, hypotension, multiple organ dysfunction syndrome (MODS), and increased mortality. Endotoxemia-derived sepsis plays an important role in sepsis syndrome, where endotoxin interacts with endothelial cells and induces detrimental effects. Nutrient consumption, including omega-3 fatty acids, ascorbic acid, and polyphenolic antioxidant flavonoids, modulates the progression of sepsis. This study aimed to investigate the impact of dietary supplementation with these nutrients on the modulation of fibrotic endothelial gene-expression patterns during sepsis and their effects on sepsis outcomes. The findings suggest that the consumption of omega-3 fatty acids and polyphenolic antioxidant flavonoids improves endotoxemia outcomes, highlighting the potential role of specific nutrient consumption in sepsis treatment.

Exploring the anticancer potential of thiadiazole derivatives of substituted thiosemicarbazones formed via copper-mediated cyclization

Summary: A series of thiosemicarbazones were synthesized and evaluated for their anticancer activity. One compound (CFM) showed potent inhibitory effects against breast adenocarcinoma and urinary bladder cancer cells, while exhibiting negligible cytotoxicity against normal cells. Molecular docking studies suggested effective binding of these compounds with cancer proteins and serum proteins. These findings provide insights into the bioactivities of thiosemicarbazones.

Sex hormones may play a protective role during SARS-CoV-2 infection, but impaired testosterone synthesis in men weakens this protective effect.

Relationships between the structural characteristic of curcumins that affect cell proliferation of hepatocarcinoma cells.

Summary: The relationship between the structural characteristic of curcumin and dimethoxycurcumin and their antitumor activity was investigated. Treatment of HepG2 cells with curcumin and dimethoxycurcumin for 24 hours induced apoptosis and inhibited cell proliferation in a dose-dependent manner. Docking and DFT calculations suggested a structure-activity relationship between the activities of dimethoxycurcumin and the structural characteristics of curcumin in inducing apoptosis in HepG2 cells.

Research has shown that an increase in inflammatory signaling pathways in patients with myeloproliferative neoplasms (MPN) leads to the differentiation of stem cells in the bone marrow into cells with dysregulated extracellular matrices, causing fibrosis. By combining two drugs that target these pathways, the development of fibrosis in MPN cell cultures can be inhibited.
