Long-standing arterial hypertension is associated with Pitx2 down-regulation in a rat model of spontaneous atrial tachyarrhythmias

Aims The timecourse of left atrial Pitx2 down-regulation in the setting of atrial tachyarrhythmias remains unknown. Accordingly, we aimed to assess the age dependency of left atrial Pitx2 expression in an experimental model of spontaneous atrial tachyarrhythmias in rats. Methods and results Atrial sampling was performed in three groups (n = 4 each) of young (14-week-old), adult (24-week-old), and ageing (48-week-old) spontaneously hypertensive rats (SHRs), in which we previously demonstrated the age dependency of spontaneous atrial tachyarrhythmias, and three groups (n = 4 each) of age-matched normotensive Wistar-Kyoto (WKY) rats. mRNA expression of Pitx2 was studied using real-time polymerase chain reaction. Ageing SHRs presented significantly lower left atrial Pitx2 expressions compared with age-matched WKY rats (P = 0.02), while no significant difference was observed between young or adult SHRs and age-matched WKY rats (both P > 0.05). Among SHRs, Pitx2 expressions showed a progressive, age-dependent decrease (34.9 +/- 6.7 in young SHRs, 17.1 +/- 3.6 in adult SHRs, and 10.7 +/- 1.7 in ageing SHRs, P = 0.04) and were significantly negatively correlated with both age (Spearman r = -0.86, P < 0.01) and heart weight (Spearman r = -0.76, P < 0.01). Conclusion The present study suggests the presence of age-dependent left atrial Pitx2 down-regulation in SHRs. The strong negative correlation between left atrial Pitx2 expression and heart weight among SHRs may indicate a link between long-standing arterial hypertension and Pitx2-related atrial arrhythmogenicity.

Factors associated with kalemia in renal disease

Association between eating behaviour and 13-year cardiovascular damages in the initially healthy STANISLAS cohort

Summary: This study aimed to assess the associations between eating behavior and cardiovascular damage or metabolic syndrome after 13 years. The results showed that emotional eating was associated with an increased risk of diastolic dysfunction after 13 years. External eating was slightly associated with lower pulse-wave velocity. These findings suggest that cardiovascular prevention should take into account eating behavior and include teaching emotion regulation skills.

Summary: This study aimed to evaluate the predictive value of TRI- SCORE for in-hospital mortality in redo ITVS patients and compare its accuracy with conventional surgical risk scores. The study found that TRI- SCORE was the only surgical risk score associated with in-hospital mortality, with an AUC value of 0.83, much higher than logistic EuroSCORE and EuroSCORE II. Therefore, TRI- SCORE can guide the clinical decision-making process.

Summary: This study aimed to characterize the outcomes of severe meningoencephalitis patients requiring intensive care. A multicenter international cohort study was conducted from 2017 to 2020 in 68 centers across 7 countries. Among the enrolled patients, 50.5% had a poor functional outcome at 3 months, and the independent factors associated with this outcome included age > 60 years, immunodepression, delayed ICU admission, low Glasgow Coma Scale score, motor deficits, respiratory and cardiovascular complications, while administration of certain medications on ICU admission was protective.

Sulforaphane exhibits potent renoprotective effects in preclinical models of kidney diseases: A systematic review and meta-analysis

Summary: This review analyzed the renoprotective effects of sulforaphane (SFN) in preclinical models of kidney diseases. The results showed that SFN improved renal function, reduced kidney injury, and decreased urinary protein excretion and kidney fibrosis. These findings suggest that SFN supplementation may be a potential therapeutic strategy for kidney disease.

High-sensitivity calcium biosensor on the mitochondrial surface reveals that IP3R channels participate in the reticular Ca2+ leak towards mitochondria

Summary: Genetically encoded biosensors based on fluorescent proteins (FPs) are widely used to monitor calcium ion (Ca2+) dynamics and sub-cellular spatial distribution in cells. The mitochondrial-associated membranes (MAMs), which connect the endoplasmic reticulum (ER) and mitochondria, have recently gained attention due to their role in calcium homeostasis and mitochondrial function. However, the sensitivity of the existing Ca2+ biosensors on the outer-mitochondrial membrane (OMM) is not sufficient to measure sub-micromolar changes in Ca2+ concentration. In this study, a new highly sensitive Ca2+ biosensor was developed and demonstrated to detect smaller differences in Ca2+ levels in or near the MAMs. Notably, the study also revealed the endogenous activity of IP3-receptors in the Ca2+ leak channel on the OMM during hypoxia or when SERCA activity is blocked.

Summary: The crucial role of angiogenesis in clot resolution and fibrothrombotic obstruction of the pulmonary arterial bed in CTEPH was validated using a rabbit model. Inhibiting intrathrombus angiogenesis resulted in elevated pulmonary arterial pressure, increased pulmonary vascular resistance, and right ventricular hypertrophy, accompanied by pulmonary vessel remodeling and inflammation.
