How Do Xanthophylls Protect Lipid Membranes from Oxidative Damage?

Summary: In this study, the antioxidant activity of carotenoids in biomembranes was investigated. The quenching activity of lutein and zeaxanthin in singlet oxygen generation was measured in lipid vesicles using a fluorescent probe and fluorescence lifetime imaging microscopy. The results showed that xanthophylls act as a barrier to the penetration of molecular oxygen into lipid membranes, especially in the 13-cis configuration. This photoisomerization of xanthophylls is a regulatory mechanism important for light filtration and photoprotection.

Do High Mental Demands at Work Protect Cognitive Health in Old Age via Hippocampal Volume? Results From a Community Sample

Summary: The study found that higher demands in language and knowledge, information processing, and creativity at work were associated with larger brain white and gray matter volume and better cognitive functioning. Higher demands in pattern detection may be linked to a brain retention effect in later life. However, there were no significant associations between hippocampal volume and mental demands at work.

These findings suggest that GABAR agonists could potentially serve as protectants for oligodendrocytes in central nervous system disorders.

Summary: Dravet syndrome is a rare and severe childhood-onset epilepsy caused by mutations in the SCN1A gene. Studies on Dravet mice showed persistent reduced function in inhibitory neurons and changes in excitatory neurons during different disease stages. This suggests a complex pathophysiological mechanism with reciprocal functional changes in both excitatory and inhibitory neurons influencing the developmental trajectory of the disease.

The study suggests that various gene dysfunctions can affect R-loop homeostasis through different mechanisms, thereby altering cell physiology.

Coordination between aminoacylation and editing to protect against proteotoxicity

Summary: Aminoacyl-tRNA synthetases (aaRSs), essential enzymes for protein synthesis, can cause neurological disorders when mutated. This study investigates the mechanism behind these mutations and found that combined defects in aminoacylation and editing result in severe proteotoxicity. The results show that aminoacylation deficiency predisposes cells to proteotoxic stress and impairs ribosome-associated quality control.

Summary: Depression is the most common psychiatric comorbidity in patients with mesial temporal lobe epilepsy (MTLE), with hippocampal sclerosis (HS) being the most frequent pathological hallmark. Although imaging studies show hippocampal volumetric reduction in patients with major depressive disorder, the exact role of the hippocampus in epilepsy with comorbid depression remains unclear. This study found no difference in hippocampal neuronal density in patients with MTLE-HS and co-occurring depression.

Summary: Well-designed placebo-controlled clinical trials are crucial for the development of new epilepsy treatments, but the design has remained unchanged for decades. Concerns have been raised by patients, clinicians, regulators, and innovators about the challenges of recruiting for trials, partly due to the static design of maintaining participants on add-on placebo for long periods of time when there are more therapy options available. Time-to-event trials, although having potential limitations, have been suggested as a promising mechanism to make trials more patient-friendly and reduce placebo exposure.

Summary: Limited guidance exists for the assessment and management of psychogenic non-epileptic seizures (PNES) in children. This study developed consensus-based recommendations through a scoping review and expert Delphi process. Video electroencephalography (vEEG) was the main method of PNES identification, and children with PNES were found to have higher rates of psychiatric disorders. Recommendations were made for comprehensive assessment, involvement of epilepsy experts in management, provision of educational materials, and screening for comorbidities.

Summary: Despite progress in the development of anti-seizure medications (ASMs), one third of people with epilepsy have drug-resistant epilepsy (DRE). The working definition of DRE, proposed by the International League Against Epilepsy (ILAE) in 2010, helped identify individuals who might benefit from presurgical evaluation early on. There is a need to revisit the current definition of DRE in order to improve research, develop new treatments, and identify drug resistance earlier.

Updated clinical recommendations for the management of tuberous sclerosis complex associated epilepsy

Summary: Children with tuberous sclerosis complex (TSC) often experience drug-resistant epilepsy, and significant progress has been made in understanding the mechanisms and management of TSC-associated epilepsy. Pre-symptomatic treatment with vigabatrin can delay seizure onset, while mTOR inhibition with everolimus and cannabidiol provide additional therapeutic options. Epilepsy surgery has shown to greatly improve seizure outcomes. Early identification of risk factors and genetic diagnosis, as well as the use of biomarkers, can contribute to the development of pre-symptomatic and disease-modifying strategies.

Uncommon use of intermittent glucose administration for infrequent non-epileptic paroxysmal events in GLUT1-DS

Summary: This article reports two cases of patients who were not following any particular diet and experienced infrequent paroxysmal non-epileptic events. Intermittent glucose intake before physical activity and at the onset of symptoms consistently improved the symptoms. The observations suggest the possibility of developing a new treatment strategy involving sustained increase in blood glucose. However, this strategy should currently be limited to a small group of GLUT1-DS patients not on a ketogenic diet.
