History, advancements, and perspective of biological research in deep-underground laboratories: A brief review

The world is entering a new era of exploring and exploiting the deep-underground space. With humans poised to reach historical depths in the use of the deep Earth, it is essential to understand the effect of the deep-underground environment on the health of humans and other living organisms. This article outlines the history and development of biological research conducted in deep-underground laboratories and provides insight into future areas of investigation. Many deep-underground laboratories have investigated the effects of reduced cosmic ray muons flux, searching for rare events such as proton decay, dark matter particles, or neutrino interactions, but few have focused on the influence of the environmental factors in the deep-underground on living organisms. Some studies revealed that prokaryote and eukaryote cells maintained in low levels of background radiation exhibited an stress response, which manifested as changes in cell growth, enzyme activity, and sensitivity to factors that cause genetic damage; however, the underlying mechanisms are unclear. There remains an urgent need to understand the detrimental and beneficial biological effects of low background radiation and other factors in the deep-underground on humans and other organisms. Consequently, a multidisciplinary approach to medical research in the deep-underground has been proposed, creating a new discipline, deep-underground medicine, and representing a historical milestone for exploring the deep Earth and in medical research.

Identification and verification of the ferroptosis- and pyroptosis-associated prognostic signature for low-grade glioma

Summary: Accumulating evidence suggests that ferroptosis and pyroptosis play important roles in the tumorigenesis of low-grade glioma (LGG). This research aimed to classify molecular subtypes of LGG and identify a novel multigene signature based on ferroptosis- and pyroptosis-related genes. The study found that the FPRGs had prognostic value and developed a six-gene prognostic signature that outperformed other recognized signatures in predicting the survival probability of LGG patients.

Comprehensive characterization of extracellular matrix-related genes in PAAD identified a novel prognostic panel related to clinical outcomes and immune microenvironment: A silico analysis with in vivo and vitro validation

Summary: The functions of ECM-related genes (ECMGs) in pancreatic adenocarcinoma (PAAD) were systematically evaluated. Seven ECMGs were identified as differentially up-regulated in PAAD and significantly linked to tumor stage and prognosis. A novel mRNA-lncRNA-based prognostic panel (ECM-APP) was developed and validated for accurately predicting clinical outcomes of PAAD patients.

TESS Timings of 31 Hot Jupiters with Ephemeris Uncertainties

Summary: A precise transit ephemeris is crucial for follow-up exoplanet observations. By comparing TESS Object of Interest (TOI) transit timings with archival ephemeris, it is found that 31 out of 262 hot Jupiters have timing offsets, with WASP-161b showing the largest offset of -203.7 +/- 4.1 minutes. These offsets have a median value of 17.8 minutes, equivalent to 3.6 sigma. Using a self-generated pipeline, TESS timings are generated for these 31 hot Jupiters in each sector, refining and updating the previous ephemeris with a median precision of 0.82 minutes until 2025 and 1.21 minutes until 2030.

A membrane-based seawater electrolyser for hydrogen generation

Summary: This article introduces a method for direct seawater electrolysis for hydrogen production, which can solve the problems of side reactions and corrosion. In the experiment, this method stably operated for over 3200 hours under practical application conditions. The method is efficient, size-flexible, scalable, and has high practical value without increasing the operation cost.

Immune-related potential biomarkers and therapeutic targets in coronary artery disease

Summary: This study aimed to identify and validate the diagnostic and therapeutic potential of differentially expressed immune-related genes (DE-IRGs) in coronary artery disease (CAD). Key marker genes were identified through functional enrichment analysis and immune cell infiltration comparison. Potential targeted drugs were also discovered. The effectiveness of the immune-related prognostic model for CAD was confirmed through validation.

Evaluation of fetal cardiac function in fetal growth restriction via fetal HQ analysis based on two-dimensional STI

Summary: This study aimed to explore the value of fetal HQ analysis based on speckle tracking imaging (STI) to evaluate the global and regional cardiac function of fetuses with early-onset or late-onset FGR. The study found that FGR affected the global and segmental cardiac function of both ventricles and altered Doppler indexes. The results showed that FAC and GLS had satisfactory repeatability in evaluating fetal cardiac function.

Correlation between pancreatic cancer and metabolic syndrome: A systematic review and meta-analysis

Summary: This meta-analysis examines the correlation between metabolic syndrome (MetS) and pancreatic cancer. The results show that MetS is strongly associated with an increased risk of developing pancreatic cancer. This association holds true regardless of gender, and hypertension, hyperglycemia, and low HDL-c levels contribute to this relationship. However, obesity and hypertriglyceridemia are not independently related to pancreatic cancer.

Network pharmacology prediction and molecular docking-based strategy to discover the potential pharmacological mechanism of action of Wang Bu Liu Xing (Semen vaccariae) for colorectal cancer

Summary: This study investigated the therapeutic effects and potential mechanisms of Wang Bu Liu Xing (SV), a traditional Chinese medicine, on colorectal cancer (CRC) using network pharmacology. The results showed that the active component swerchirin in SV may inhibit CRC by interacting with key targets such as BCL2L1, CDK2, and SERPINE1, and the p53 signaling pathway may be involved in the anti-CRC effect of SV. Molecular docking confirmed the strong binding between swerchirin and its target protein. This study offers a promising approach for understanding the therapeutic pathways and identifying molecules in traditional Chinese medicine.

Identification of lipid metabolism-related biomarkers for diagnosis and molecular classification of atherosclerosis

Summary: This study aimed to investigate lipid metabolism-related molecular clusters and develop a diagnostic model for atherosclerosis. A total of 29 differentially expressed lipid metabolism-related genes were identified, primarily involved in cholesterol and lipid metabolism, the PPAR signaling pathway, and regulation of the inflammatory response. A three-gene diagnostic model (ADCY7, SCD, and CD36) was constructed, showing good predictive performance and association with immune cell infiltration.

Identification and immunological characterization of lipid metabolism-related molecular clusters in nonalcoholic fatty liver disease

Summary: This study explored lipid metabolism-related clusters in NAFLD and established a prediction biomarker. Dysregulated lipid metabolism-related genes and immune responses were identified in NAFLD. A machine learning model, specifically a random forest model, showed good predictive performance. The final five-gene random forest model exhibited excellent predictive power in two test sets.

Unveiling the advantages of an ultrathin N-doped carbon shell on self-supported tungsten phosphide nanowire arrays for the hydrogen evolution reaction experimentally and theoretically

Summary: Packaging electrocatalysts with carbon shells can enhance the stability and efficiency of hydrogen evolution reaction (HER) materials. An ultrathin N-doped carbon-coated self-supported WP nanowire array (WP@NC NA) hybrid has been synthesized, which exhibits improved physicochemical stability, active sites, and conductivity. The carbon shell optimizes various steps in both acidic and alkaline HER reactions. These findings provide scientific guidance for the packaging design of carbon-encapsulating self-supported electrocatalysts.
