This work assesses the urinary metabolite signature of prematurity in newborns by nuclear magnetic resonance (NMR) spectroscopy, while establishing the role of possible confounders and signature specificity, through comparison to other disorders. Gender and delivery mode are shown to impact importantly on newborn urine composition, their analysis pointing out at specific metabolite variations requiring consideration in unmatched subject groups. Premature newborns are, however, characterized by a stronger signature of varying metabolites, suggestive of disturbances in nucleotide metabolism, lung surfactants biosynthesis and renal function, along with enhancement of tricarboxylic acid (TCA) cycle activity, fatty acids oxidation, and oxidative stress. Comparison with other abnormal conditions (respiratory depression episode, large for gestational age, malformations, jaundice and premature rupture of membranes) reveals that such signature seems to be largely specific of preterm newborns, showing that NMR metabolomics can retrieve particular disorder effects, as well as general stress effects. These results provide valuable novel information on the metabolic impact of prematurity, contributing to the better understanding of its effects on the newborn's state of health.

Zinc Prevents DNA Damage in Normal Cells but Shows Genotoxic and Cytotoxic Effects in Acute Myeloid Leukemia Cells

Maria Ines Costa, Beatriz Santos Lapa, Joana Jorge, Raquel Alves, Isabel Marques Carreira, Ana Bela Sarmento-Ribeiro, Ana Cristina Goncalves

Summary: Zinc plays a dual role in modulating the DNA damage response in acute myeloid leukemia (AML), increasing genotoxicity and cytotoxicity in leukemia cells while preventing damage accumulation in normal cells.

Summary: This study aimed to determine the predictors of early recurrence (ER) and late recurrence (LR) after catheter ablation (CA) for atrial fibrillation (AF) and establish the risk of LR in patients who experience ER. The results showed that the risk of LR after CA was significantly higher in patients with ER (five-fold increased risk). These findings highlight the importance of clarifying the clinical role of the blanking period.

Quality Indicators Compliance and Survival Outcomes in Breast Cancer according to Age in a Certified Center

Fernando Osorio, Antonio S. Barros, Barbara Peleteiro, Isabel Amendoeira, Jose Luis Fougo

Summary: In this study, we compared the outcomes of breast cancer among three age groups and found that older patients were more susceptible to suboptimal treatment and had a lower overall survival rate. This was not associated with the cancer itself, but rather with undertreatment. The study suggests that tumor characteristics and treatment compliance are better predictors of survival than chronological age.

The impact of heart failure therapy in patients with mildly reduced ejection fraction: a network meta-analysis

Marta Leite, Francisco Sampaio, Francisca A. Saraiva, Silvia O. Diaz, Antonio S. Barros, Ricardo Fontes-Carvalho

Summary: Recently, HF guidelines have re-classified HF patients with LVEF between 41% and 49% as HFmrEF. The treatment of HFmrEF patients is often considered a grey zone due to the lack of RCTs exclusively conducted on this population. A network meta-analysis was conducted to compare the treatment effect of different medications in HFmrEF cardiovascular outcomes.

Cardiovascular risk factors during pregnancy impact the postpartum cardiac and vascular reverse remodeling

Ana Filipa Ferreira, Maria Joao Azevedo, Juliana Morais, Fabio Trindade, Francisca Saraiva, Silvia Oliveira Diaz, Ines Nuno Alves, Mariana Fragao-Marques, Carla Sousa, Ana Paula Machado, Adelino Leite-Moreira, Benedita Sampaio-Maia, Carla Ramalho, Antonio Sousa Barros, Ines Falcao-Marques

Summary: Pregnant women with cardiovascular risk factors are at high risk of developing cardiovascular disease later in life. Our study found that pregnant women undergo cardiovascular remodeling during pregnancy, which reverses after delivery. Arterial hypertension, smoking, and obesity are independent predictors of increased left ventricular mass. Additionally, circulating biomarkers such as C-reactive protein and ST2/IL33 receptors may be associated with worse cardiac remodeling.

Impact of Sea Warming and 17-a-Ethinylestradiol Exposure on the Lipid Metabolism of Ruditapes philippinarum Clams

Joao A. Rodrigues, Daniela S. C. Bispo, Monica G. Silva, Rita Araujo, Amadeu M. V. M. Soares, Rosa Freitas, Ana M. Gil

Summary: This study investigates the effects of exposure to 17-α-ethinylestradiol (EE2) on lipophilic extracts of Ruditapes philippinarum clams at 17 degrees C and 21 degrees C. The results show that EE2 exposure at 17 degrees C leads to a slight increase in membrane rigidity at low concentrations, while at higher concentrations, lipid metabolism stabilizes. In contrast, at 21 degrees C, lipid metabolism responds to EE2 at 125 ng/L, with the antioxidant docosahexaenoic acid (DHA) helping to mitigate oxidative stress. The study also identifies new potential markers of stress mitigation, such as high levels of phosphatidylcholine (PtdCho), polyunsaturated fatty acids (PUFAs), and linoleic acid, as well as low PUFA/glycine ratios.

Tumor Lipid Signatures Are Descriptive of Acquisition of Therapy Resistance in an Endocrine-Related Breast Cancer Mouse Model

Rita Araujo, Victoria Fabris, Caroline A. Lamb, Andres Elia, Claudia Lanari, Luisa A. Helguero, Ana M. Gil

Summary: The lipid metabolism adaptations of estrogen and progesteronereceptor-positive breast cancer tumors in relation to HD to HI tumor growth transition and HIR tumor acquisition of ET resistance were investigated. Results showed increased nonesterified cholesterol forms and phospholipids, and decreased triglycerides and fatty acids during tumor progression. These changes suggest a shift toward more activated cholesterol synthesis, β-oxidation, and phospholipid biosynthesis to sustain tumor growth. Certain compound levels and ratios were identified as potential markers for HD-HI-HIR progression.

The Association between Gestational Diabetes and the Microbiome: A Systematic Review and Meta-Analysis

Rita Almeida Teixeira, Claudia Silva, Antonio Carlos Ferreira, Diana Martins, Adelino Leite-Moreira, Isabel M. Miranda, Antonio S. Barros

Summary: Gestational diabetes, affecting 10% of pregnancies, has implications for both mother and child. This study reviewed the literature on the differences in microbiota composition between pregnant women with gestational diabetes and healthy pregnancies. The findings suggest that certain bacteria are more abundant in healthy pregnancies, while others are more abundant in gestational diabetes. The study also highlighted the potential of microbiome-targeted therapies for gestational diabetes.
