Summary: Tobacco use, but not alcohol drinking, significantly increases the risk of frailty and falling, according to a two-sample Mendelian randomisation analysis using genetic instruments. Genetically determined initiation of smoking and cigarettes consumption per day were both positively associated with frailty and falling, while there was no significant causal association between alcohol use and the risk of frailty and falling.

Summary: A case-control study conducted in Singapore found that family history is a significant risk factor for lung cancer in Singaporean Chinese women, especially among never-smokers. This highlights the importance of considering family history in assessing lung cancer risk, particularly in non-smoking populations.

Summary: Through analysis of a tuberculosis outbreak and molecular characteristics of the MtZ strain, several SNPs in genes related to virulence, pathogenesis, and survival, as well as other genomic polymorphisms, were identified, which may contribute to its success in transmission among the population.

Summary: Previous tuberculosis disease is an important and under-recognized risk factor for COPD and poor lung function in low-income and middle-income countries. Better tuberculosis control is likely to reduce the global burden of COPD.

Summary: In a community-wide TB screening programme in Karachi, Pakistan, gender-based differences were observed, with female attendees less likely to undergo sputum collection and testing as well as initiate treatment, despite having a higher age-standardised prevalence of active TB. Integrating gender into the design and monitoring of TB screening programmes is crucial to ensure equal benefits for women and men.

Summary: This study conducted a systematic review and meta-analysis comparing high-dose rifamycin (HDR) regimens to standard-dose rifamycin (SDR) regimens in the treatment of tuberculosis. The results showed no significant difference in severe adverse events, death rate, 2-month culture conversion, and relapse between the two regimens. Further studies are needed to identify specific groups that may benefit from HDR.

Summary: Among children aged 6-10 years, living with patients with clinical or subclinical tuberculosis increases the risk of tuberculin skin test positivity. The transmission of subclinical tuberculosis is still inconclusive. The findings support the hypothesis that smear-positive subclinical tuberculosis contributes to Mycobacterium tuberculosis transmission.

Summary: This study aims to assess the incidence and predictors of acquired resistance to second-line antituberculosis drugs (SLDs) during multi-drug resistant tuberculosis (MDR-TB) treatment. The study will systematically search and analyze published articles up to March 2023, and will use statistical software to estimate the pooled incidence and effect measures of acquired resistance. The findings will be disseminated through peer-reviewed journals and scientific conferences.

Summary: This study found that exposure to second-hand smoke (SHS) is associated with an increased risk of tuberculosis (TB) and latent TB infection (LTBI) in children. Furthermore, smoking also affects the accuracy of Interferon-Gamma Release Assays (IGRAs) and cytokine immune responses.

Summary: The genetic diversity of Mycobacterium tuberculosis plays a role in disease severity and transmissibility. In this study, the phenotyping of M. tuberculosis from a persistent outbreak in the UK revealed higher lipid contents and more hydrophobic cell surfaces compared to nonoutbreak isolates. Deletions in specific genes were also found in the outbreak isolates, potentially altering the interactions between the bacteria and immune cells. Reintroduction of one of these genes led to a phenotype resembling nonoutbreak strains. Understanding the microbiological characteristics and genetic polymorphisms associated with outbreaks can inform tuberculosis control strategies.

Summary: The study found that education level was associated with the prevalence of smoking, regardless of social class, and specific behaviors related to same-sex sexual experiences were associated with a higher prevalence of smoking. Regular alcohol consumption was also correlated with a higher prevalence of cigarette smoking.

Summary: This study aimed to characterize the lineages of Mycobacterium tuberculosis complex causing childhood pulmonary tuberculosis and evaluate drug sensitivity patterns. The majority of isolates belonged to the Euro-American lineage, and drug resistance was low. A large-scale prospective study is recommended to fully understand the molecular epidemiology of MTBC lineages in children.

Summary: The study evaluated the clinical and antibiotic resistance impact of carbapenems stewardship programs, demonstrating that interventions on prescription for patients without justified carbapenem treatment could improve prescription accuracy. Over five years, there was a decrease in carbapenem consumption and rates of hospital-acquired MDR BSIs, as well as an increase in the consumption of other antibiotics, overall leading to a reduction in inpatient days and candidemia.
