Summary: The study found that N-2-Hydroxypropyl trimethyl ammonium chloride chitosan/N, O-carboxymethyl chitosan nanoparticles (N-2-HACC/CMCS NPs) showed good immunogenicity and no toxicity as a universal vaccine adjuvant/delivery system. The prepared N-2-HACC/OVA/CMCS NPs induced higher levels of antibodies and immune responses in mice, suggesting their potential in enhancing humoral, cellular, and mucosal immune responses.

Summary: In this study, N-trimethyl chitosan (TMC) was used as a mucoadhesive adjuvant to enhance the oral bioavailability and antitumour effects of gemcitabine formulated into nanocomplexes. The results showed that the TMC-modified nanocomplexes significantly improved the oral drug bioavailability and exhibited potent anticancer efficacy.

Summary: This report presents the encapsulation of amoxicillin in N-2-Hydroxypropyl trimethyl ammonium chloride chitosan and N,O-carboxymethyl chitosan nanoparticles for biomedical applications. The synthesized nanoparticles demonstrated sustained drug release, good cytocompatibility and broad-spectrum antimicrobial activity.

Summary: This study investigates the property and mechanism of intestinal absorption of nanoparticles coated with trimethyl chitosan derivatives (PPTT-NPs), which have previously been shown to enhance the oral bioavailability of panax notoginseng saponins. The results demonstrate that TMC-VB12 and TMC-Cys effectively promote the transport of PPTT-NPs, increasing their adhesion and permeation in the intestines. These findings suggest the potential for oral delivery of BCS III drugs.

Summary: A two-step method was used to synthesize N,N,N-trimethyl chitosan (TMC) from chitosan, with molecular weight ranging from 29 to 136 kg/mol and quaternization degree from 70% to 82%. Size exclusion chromatography (SEC) coupled with multiangle laser light scattering was employed to analyze the molecular weight and chain conformation, showing a significant increase in chain flexibility upon N-methylation, possibly due to the breakage of intramolecular hydrogen bonds between chitosan repeating units.

Dynamic viscoelasticity tests show that HACC exhibits both viscous behavior similar to a fluid and elastic properties of weak gel.

Summary: This study aimed to enhance the oral absorption of Panax notoginseng saponins (PNS) through nanoparticles modified with thiolated trimethyl chitosan (TMC-Cys) and wheat germ agglutinin (WGA). The results showed that the modified nanoparticles exhibited delayed release, tolerance to gastric acids and digestive enzymes, as well as efficient cellular uptake and transport capabilities. Animal experiments demonstrated that the modified nanoparticles effectively overcame the mucus layer barrier and increased the permeability coefficients of PNS in the small intestine.

Summary: This work proposes a modified method using microfluidics combined with the microemulsion technique to manufacture chitosan nanoparticles. The synthesized nanoparticles showed improved size control and distribution compared to batch synthesis. The total encapsulation of IgG protein was confirmed during the fabrication of chitosan/IgG-loaded nanoparticles. The proposed materials have the potential to be used as carrier-delivery systems.

Summary: The study developed a VP2 protein subunit vaccine against Porcine parvovirus (PPV) using N-2-Hydroxypropyl trimethyl ammonium chloride chitosan (N-2-HACC) as the adjuvant, showing that the vaccine induced higher antibody levels and long-term immune protection in immunized sows. However, the vaccine mainly stimulated humoral immunity rather than cellular immunity, and provided 100% protective efficacy against PPV infection in sows. This suggests that N-2-HACC could be a potential adjuvant for controlling PPV.

Summary: Antimicrobial hydrogels are a major focus for repairing soft tissues and preventing bacterial infections, with new hydrogels designed in this study showing high biocompatibility through physical crosslinking reactions. The hydrogels exhibit significant antibacterial activity against Staphylococcus aureus and Escherichia coli, with those containing high TMC content showing superior inhibition zones compared to pure TMC. Additionally, the cytotoxicity testing on normal cell lines VERO and lung (Wi38) cells demonstrates high cell viability at lower concentrations.

Summary: This article discusses the effect of N,N,N-trimethyl chitosan (TMC) on a mimetic pulmonary surfactant, Curosurf (R). The results show that TMC has a strong interaction with Curosurf (R), leading to the formation of aggregates at electrostatic charge stoichiometry. At the nanoscale, Curosurf (R) undergoes a profound reorganization of its lipid vesicles in terms of size and lamellarity. The neutralization of cationic charges by pulmonary surfactant may inhibit the permeation enhancement capacity of TMC, especially at low TMC content. Therefore, the permeation properties of pulmonary surfactant-neutralized TMC should be evaluated for its applicability as a permeation enhancer for inhalation in the alveolar region.
