Gastro-esophageal reflux disease (GERD) is the most common digestive disease in industrialized countries (Europe and North America) and is associated with microscopic changes in the squamous epithelium. However, biopsy is not presently included in the routine diagnostic flow chart of GERD. In contrast, esophageal biopsy is mandatory when diagnosing Barrett's esophagus. High quality histology reports are necessary to provide information on diagnosis and can also be important for research and epidemiological studies. It has been evident for decades that pathology reports vary between institutions and even within a single institution. Standardization of reporting is the best way to ensure that information necessary for patient management is included in pathology reports. This paper details the histological criteria for diagnosing GERD-associated microscopic esophagitis, other forms of esophagitis with specific features and columnar metaplasia in the lower esophagus (Barrett's esophagus). It provides a detailed description of appropriate sampling criteria, individual lesions and how they contribute to the histology report. (C) 2011 Editrice Gastroenterologica Italiana S.r.l. Dasatinib-induced Crohn's-like colitis

Federica Grillo, Luca Carlin, Andrea Remo, Matteo Fassan, Claudia Mescoli, Michela Campora, Luigi Caserta, Fabrizio Mazza, Luca Mastracci

Summary: Dasatinib-induced colitis may present histological features similar to Crohn's disease, including microgranulomas and active inflammation, and complete remission can be achieved after treatment interruption.

Counting mitoses in gastrointestinal stromal tumours (GISTs): variable practices in the real-world setting and their clinical implications

Michela Campora, Michele Paudice, Alessandro Gambella, Danila Comandini, Paola Parente, Marta Sbaraglia, Angelo Paolo Dei Tos, Federica Grillo, Luca Mastracci

Summary: Mitotic count is an important prognostic indicator in gastrointestinal stromal tumours. The study found that many pathologists still use 50 high-power fields instead of 5 mm^2 for mitotic count evaluation, leading to non-standardized assessment and risk stratification errors.

Evaluating mismatch repair deficiency in colorectal cancer biopsy specimens

Summary: This study aims to identify the advantages, disadvantages, and potential pitfalls of MMR evaluation on biopsy tissue for colorectal cancer. The results show that biopsy specimens often have indeterminate stains, particularly for MLH1. However, this issue can be solved by reducing primary incubation times, allowing for accurate assessment of MMR status.

Gut inflammation and tumorigenesis: every site has a different tale to tell

Alessandro Vanoli, Paola Parente, Matteo Fassan, Luca Mastracci, Federica Grillo

Summary: Gut inflammation disrupts gastrointestinal homeostasis and is associated with cancerogenesis. Various chronic inflammatory disorders of the tubular gastrointestinal tract have been linked to an increased risk of neoplasms, including epithelial tumors (esophageal, gastric, small bowel, and colorectal) and lymphomas. Studies have investigated the mechanisms and changes in the microenvironment and microbiome that lead to genetic and/or epigenetic alterations and eventually tumorigenesis, typically following an inflammation-dysplasia-carcinoma sequence.

Summary: This study conducted genomic analysis on a neoadjuvant chemotherapy-resistant gastroesophageal tumor and identified a novel gene fusion, MSI2-C17orf64, that has never been reported before. This finding highlights the potential of MSI2 as a therapeutic target.

Summary: Esophageal adenocarcinoma (EAC) is a severe malignancy with increasing incidence, poorly understood pathogenesis, and low survival rates. TP53 gene alterations, especially missense mutations, negatively affect cancer-specific survival in EAC. HNF1alpha gene disruption is associated with other gene alterations and is identified as a new EAC-mutated gene.

Single-cell proteo-genomic reveals a comprehensive map of centrosome-associated spliceosome components

Luigi Cerulo, Nunziana Pezzella, Francesca Pia Caruso, Paola Parente, Andrea Remo, Guido Giordano, Nicola Forte, Johan Busselez, Federico Boschi, Mirco Galie, Brunella Franco, Massimo Pancione

Summary: Ribonucleoprotein (RNP) condensates play a crucial role in controlling RNA metabolism and splicing events in animal cells. By using spatial proteomics and transcriptomics, researchers have identified the RNP interaction networks at the centrosome, which is the main microtubule-organizing center in animal cells. They found that specific cell-type centrosome-associated spliceosome interactions are localized in subcellular structures involved in nuclear division and ciliogenesis. This research provides insights into the functions of cytoplasmic condensates in defining cell identity and the origin of rare diseases.
