Weight loss interventions and nonalcoholic fatty liver disease: Optimizing liver outcomes

Summary: The increase in prevalence of obesity, type 2 diabetes, and nonalcoholic fatty liver disease (NAFLD) presents a significant global health challenge. Weight loss has been identified as the most effective treatment for NAFLD, improving biomarkers, preventing progression, and sometimes reversing fibrosis. Pharmacotherapy, either alone or in combination with lifestyle interventions, is bridging the efficacy gap between diet and exercise and weight-loss surgery in reversing obesity.

Summary: There is a growing need for more effective treatment of obesity and its complications, with current medications falling short of the efficacy seen with bariatric surgery. Gut peptide analogues are leading the way in clinical development and are likely to meet regulatory requirements soon. Early-stage drug development targeting different energy balance pathways may lead to more complex and personalized treatment guidelines.

Prevalence of Baseline Cardiac Arrhythmias in Participants with Overweight or Obesity in Phase 1 Clinical Trials: Analysis of 24-Hour Holter Electrocardiogram Recordings

Summary: Although prevalence of cardiac arrhythmias in healthy volunteers has been estimated, there is limited baseline data for populations such as overweight and obese individuals, who are increasingly participating in clinical trials. This study examined the baseline prevalence of arrhythmias in overweight or obese participants in two phase 1 trials of weight management medications. The findings showed that most arrhythmias were rare, and the prevalence mirrored that of healthy participants with normal weight. Knowledge of the baseline prevalence of arrhythmias in overweight and obese individuals can inform trial eligibility criteria and improve decision-making in weight management clinical trials.

Body mass index trajectories among people with obesity and association with mortality: Evidence from a large Israeli database

Summary: This study examined BMI trajectories over 5 years in a large population with obesity and found that individuals with stable high weight had the lowest mortality. These findings contribute to the scientific understanding of the impact of weight trajectories on health outcomes.
