Summary: This study utilized human induced pluripotent stem cells to generate cerebral organoids carrying a KCNJ11 mutation, demonstrating the impact of the mutation on neural network development and function. Differential analysis revealed a decrease in neuron number and reduced neural network synchronization due to the mutation, with partial improvement observed with continuous treatment of the K-ATP blocker.

Summary: In recent years, there has been a significant improvement in the prognosis of patients with Hodgkin lymphoma (HL). However, the study found that HL and its treatment can have an impact on the fertility of male patients, with hematopoietic stem cell transplantation (HSCT) associated with severe impairment of sperm motility. First-line treatment, on the other hand, showed a statistically significant improvement in fertility in terms of motility for HL patients who did not undergo HSCT.

Summary: This study reports the underlying genetic variants of a newly described distinct phenotype of newborn glaucoma called neonatal-onset congenital ectropion uveae (NO-CEU). Through clinical exome sequencing of 13 children with NO-CEU, the study identified CYP1B1 variants associated with the phenotype. The findings also showed that different genotypes have an impact on ocular manifestations and visual outcomes in these patients.

Summary: The current state of wellness in medicine is characterized by narrow conceptualizations and solutions informed by a disease-focused approach. Moving forward requires clear definitions of wellness, methodologies incorporating direct interaction with participants, and a shift away from disease-model approaches. Wellness should be reimagined as healthy and authentic engagement with the inevitable challenges of a medical career to support the development of coping mechanisms and resilience.

Summary: This study aimed to assess the myelination pattern in neonates with neonatal encephalopathy (NE) who developed brain injury compared to those without injury over the first month of life. The results showed that in NE neonates treated with hypothermia who developed brain injury, birth asphyxia impaired myelination in various brain regions, indicating a potential link between birth asphyxia and myelination deficits.

Summary: Perinatal hypoxia-ischemia is the most common cause of acute neonatal brain injury, and inflammation plays a significant role in its pathogenesis. Recent studies have discovered that lactate can regulate inflammation through histone lactylation. Understanding the interaction between histone lactylation and inflammation could lead to the development of immunomodulatory therapies for neonatal brain damage.

Summary: The study found that CLN7 may be involved in the infection process of the SARS-CoV-2 virus. CLN7 deficiency leads to a significant reduction in viral load, while overexpression of CLN7 increases viral load. This research provides a potential target for drug development against COVID-19 and helps us understand the physiological function of CLN7.

Summary: This study explored the effect of thiamine on the prognosis of patients with sepsis. The results showed that patients who received thiamine treatment had a significantly lower ICU mortality risk. These findings suggest a beneficial effect of thiamine supplementation on the prognosis of sepsis patients. Further randomized controlled trials are needed to confirm its effectiveness.

Summary: This study investigated the performance of dyslexics compared to controls in estimating and comparing time intervals defined by visual stimuli. The results showed that dyslexics had poorer accuracy in visual-based duration perception. While it is unclear to what extent visual deficits contribute to their difficulties in judging time intervals, the findings suggest that attention and/or engagement may play a role in dyslexics' timing performance.

Summary: Obesity leads to physical comorbidities and mental health consequences. This study investigates the effect of physical activity on psychological well-being through the brain-gut microbiome (BGM) system in individuals with high BMI. The results show that higher physical activity is associated with enhanced connectivity in appetite control brain regions, while lower physical activity is linked to increased connections in the emotional regulation network. Higher physical activity is also associated with microbiome and metabolite signatures that protect against mental health and metabolic disorders. These findings highlight the psychological and resilience benefits of physical activity, beyond its metabolic effects, which may be attributed to BGM interactions.

Summary: The study aims to characterize the sleep phenotypes of individuals with Rett Syndrome. The results show differences in sleep latency, total sleep time, and wake time after sleep onset among patients with different levels of clinical severity, particularly in hand functioning and walking. These differences are associated with increased deep sleep and reduced rapid eye movement sleep, which may be linked to (psycho)motor impairment in patients with MECP2 mutations.

Summary: This study aimed to investigate the factors associated with sleep quality in healthcare students and determine whether depressive and anxiety symptoms can explain some of the associations between sleep quality and self-rated health. Structural equation modeling revealed that depressive and anxiety symptoms partially explain the association between sleep quality and self-rated health.

Summary: The study developed and validated a machine learning algorithm for predicting moderate to severe obstructive sleep apnea syndrome (OSAS) in otherwise healthy children. The algorithm utilized multivariable logistic regression and the cforest algorithm to analyze a large cross-sectional dataset of children with sleep-disordered breathing. The findings demonstrated that a machine learning algorithm using the cforest algorithm and predictors associated with OSAS can accurately predict moderate to severe OSAS in mostly obese but otherwise healthy children.

Summary: This systematic review evaluates the association between attention deficit hyperactivity disorder (ADHD) and restless legs syndrome (RLS) based on PRISMA guidelines. After searching databases for relevant articles, 29 were included out of the 208 identified between 1996 and 2022. The findings suggest a strong co-occurrence between RLS and ADHD, and highlight sleep fragmentation as a common characteristic.

Summary: Rett syndrome is a neurodevelopmental disorder characterized by the loss of spoken language and apraxia. A systematic review of 12 studies revealed poor non-verbal social functioning in individuals with Rett syndrome, particularly in sustained attention and recognition of novel social stimuli. However, they showed similar responsiveness to social stimuli as typically developing individuals. Music therapy has been found to improve their skills. The caregiver's perception and familiar situations may influence the interpretation of their brief gaze intent.

Summary: This study assessed the personality characteristics of children and adolescents with anxiety disorder from a maternal perspective. The results showed that patients in the clinical group exhibited higher rates of internalizing symptoms, less interest in hobbies, less adherence to social organizations, impairment in social activities, and commitment to school performance compared to the control group. It was also found that there was a positive correlation between the mothers' symptoms and the characteristics of anxiety in their children.

Summary: This study aimed to assess the acceptability of a serious game prototype for gaze stabilization training in children with vestibular hypofunction (VH) and analyze its effects on dynamic visual acuity (DVA) and motor performance. The results showed that the game training was well-tolerated and led to improvements in DVA and motor performance. This suggests that serious game-based vestibular rehabilitation could be a promising approach for children with VH in the future.
