Characterizations of two grass carp Ctenopharyngodon idella HMGB2 genes and potential roles in innate immunity

High-mobility group box 2 (HMGB2) protein is a chromatin-associated nonhistone protein, involved in transcriptional regulation and nucleic-acid-mediated innate immune responses in mammalian. However, the function of piscine HMGB2 in innate immune responses is still unknown. In the present study, two HMGB2 homologue genes (CiHMGB2a, CiHMGB2b) were identified and characterized in grass carp (Ctenophaungodon idella). Both CiHMGB2a and CiHMGB2b genes encode proteins with 213 amino acids, sharing 71.4% identities and containing two basic HMG boxes and an acidic tail. The deduced protein sequences showed the most identities to HMGB2a (93%) and HMGB2b (86.4%) of zebrafish (Danio rerio), respectively. Quantitative real-time RT-PCR (qRT-PCR) analysis showed that CiHMGB2a and CiHMGB2b were constitutively expressed in all the 15 tested tissues. Post grass carp reovirus (GCRV) infection, mRNA levels of CiHMGB2a and CiHMGB2b were strongly up-regulated in spleen and head kidney and mildly modulated in C idella kidney (CIK) cells. Meanwhile, mRNA expressions of CiHMGB2a and CiHMGB2b were significantly regulated by viral pathogen associated molecular patterns (PAMPs) polyinosinic-polycytidylic potassium salt (poly(I:C)) and bacterial PAMPs lipopolysaccharide (LPS), peptidoglycan (PGN) challenge in CIK cells. In CiHMGB2a and CiHMGB2b over-expression cells, expressions of CiHMGB2a and CiHMGB2b facilitated each other; transcription levels of CiTRIF, CiMyD88, CiIPS-1 and CiMx1 were remarkably enhanced, whereas CiIFN-I was inhibited, compared with those in cells transfected with pCMV (control plasmid); after GCRV challenge, all those tested genes were up-regulated with divergent expression profiles. Antiviral activities of CiHMGB2a and CiHMGB2b were manifested by the delayed appearance of cytopathic effect (CPE) and inhibition of GCRV yield. All those results demonstrate that CiHMGB2a and CiHMGB2b not only mediate antiviral immune responses but also involve in responding to viral/bacterial PAMPs challenge, which provides novel insights into the essential role of HMGB2 in innate immunity. Cyprinid-specific duplicated membrane TLR5 senses dsRNA as functional homodimeric receptors

Summary: The membrane-bound Toll-like receptor 5 (TLR5) in cyprinid grass carp plays a role in detecting bacterial flagellin and dsRNA analog poly(I:C), regulating interferon response and antiviral defense. Two TLR5 paralogs, CiTLR5a and CiTLR5b, have opposite effects on virus replication. The ectodomain of CiTLR5 is essential for dsRNA-induced interferon signaling, and specific glycans affect dsRNA-IFN signaling but not flagellin-induced NF-kappa B induction. This study reveals a neofunctionalization of TLR5 in cyprinid, bridging viral and bacterial immune responses.

Summary: In this study, a recombinant Lactobacillus casei expressing multiple fragments of the key outer capsid protein of grass carp reovirus (GCRV) was constructed, and an oral microcapsule live vaccine (SCL-56310-500) was prepared. The results showed that this vaccine effectively enhanced the immune response and suppressed GCRV replication in fish, leading to a significantly higher survival rate.

Summary: This review discusses the unique biological effects of IL-2 family cytokines in regulating differentiation, survival, and activation of multiple lymphocyte lineages, as well as their expression profiles, receptors, signaling transductions, and functions in teleosts. The IL-2 family cytokines in teleosts show relatively conserved functions in lymphocyte proliferation, differentiation, development, and inflammation, similar to their counterparts in mammals.

Summary: This study evaluates the feasibility and biological activity of carboxymethyl chitosan (CMCS) loaded with Ctenopharyngodon idella interferon-gamma 2 (CiIFN-gamma 2), and examines the resistance to Aeromonas hydrophila infection in grass carp. The results show that CMCS-CiIFN-gamma 2 nanoparticles can enhance the expression of pro-inflammatory cytokines and the effective uptake of the protein, leading to improved immune protection against bacterial diseases in aquaculture.

Chitooligosaccharide boosts the immunity of immunosuppressed blunt snout bream against bacterial infections

Summary: This study reveals that chitooligosaccharide (COS) has the ability to reverse cortisol-induced immunosuppression in fish and enhance the immune activity of macrophages. Oral administration of COS directly absorbed through the intestine significantly improves the innate immunity of blunt snout bream, leading to increased survival and reduced tissue damage. Thus, COS offers potential strategies for immunosuppression prevention and control in fish.

VP4/VP56/VP35 Virus-like Particles Effectively Protect Grass Carp (Ctenopharyngodon idella) against GCRV-II Infection

Summary: This study developed a prophylactic grass carp reovirus (GCRV) vaccine using virus-like particle (VLP) assembly technology, and the highly immunogenic candidates in the vaccine were VP4, VP56, and VP35. Expression of VP4, VP56, and VP35 in both Escherichia coli and Pichia pastoris systems resulted in successful assembly of uniform, stable, and non-toxic VP4/VP56/VP35 VLPs. Vaccination with P. pastoris VLPs significantly improved survival rate, IgM levels, tissue viral load, and antioxidant immune-related indicators in grass carp.

Temperature-regulated type II grass carp reovirus establishes latent infection in Ctenopharyngodon idella brain

Summary: This study investigated the latent infection of GCRV-II in asymptomatic grass carp with GCRV infection or exposure history. It was found that GCRV-II was detectable only in the brain of grass carp during latent infection, unlike the multi-tissue distribution observed in natural infection. Additionally, GCRV-II distribution in grass carp was notably affected by ambient temperature. This study provides insights into the mechanisms of GCRV-II latent infection and reactivation, contributing to the prevention and control of GCRV pandemics.

Vitamin D3 can effectively and rapidly clear largemouth bass ranavirus by immunoregulation

Summary: This study aimed to investigate the potential effectiveness of VD3 in treating Largemouth bass ranavirus (LMBV). The results showed that feeding Largemouth bass with 15000 IU/Kg and 20000 IU/Kg of VD3 significantly suppressed and cleared LMBV pathogens. In addition, VD3 treatment increased the expression levels of key immune-related genes and enhanced antioxidant and immune activities in the serum. Tissue pathology sections also revealed a dose-dependent relationship between VD3 supplementation and tissue damage.

Mechanisms of type I interferon production by chicken TLR21

Distribution and spatiotemporal development of organised lymphoid tissues in the chicken intestinal tract

Summary: Chickens have a unique immune structure characterized by the absence of draining lymph nodes and the presence of well-developed mucosa-associated lymphoid tissue. The structure and development of chicken lymphoid tissues in the intestine, particularly Peyer's patches, have been poorly studied. However, the use of CSF1R-eGFP reporter transgenic chickens has allowed for the visualization and investigation of the development and distribution of these tissues. This research provides insights that could benefit mucosal vaccination strategies and the development of targeted vaccine delivery systems.

The NACHT-LRR domain is the functional domain of rNLRC3. These findings provide insights into the important role of rNLRC3 in combating P. multocida infection.

A novel DM9-containing protein 7 involved in regulating the expression of CgMyD88 and CgIL-17 in oyster Crassostrea gigas

CgDM9CP-7 binds to various pathogens and exhibits antimicrobial activity, while also regulating the expression of CgMyD88 and CgIL-17.

Histopathological observations and comparative transcriptome analysis of Ophiocordyceps sinensis infection of Hepialus xiaojinensis in the early stage
