Members of the ADAM (a disintegrin and metalloprotease) family are type I transmembrane proteins involved in biological processes of proteolysis, cell adhesion, cellmatrix interaction, as well as in the intracellular signaling transduction. In the present study, expression patterns of seven members of the ADAM family were investigated at the early stages of the developing cochlea by in situ hybridization. The results show that each individual ADAM is expressed and regulated in the early developing cochlea. ADAM9, ADAM10, ADAM17, and ADAM23 are initially and widely expressed in the otic vesicle at embryonic day 2.5 (E2.5) and in the differential elements of the cochlear duct at E9, while ADAM12 is expressed in acoustic ganglion cells at E7. ADAM22 is detectable in cochlear ganglion cells as early as from E4 and in the basilar papilla from E7. Therefore, the present study extends our previous results and suggests that ADAMs also play a role in the early cochlear development.

Summary: The Sonic hedgehog (Shh) signaling pathway plays crucial roles in the development of the central nervous system (CNS) and related diseases. Abnormal signaling can lead to the occurrence of neurological disorders. Studies have classified the Shh signaling pathway into canonical and non-canonical pathways.

Summary: This study aimed to establish a stable animal model of intrauterine adhesion (IUA) using a minimally invasive method that recapitulates the clinicopathologic characteristics of IUA. The results showed a positive correlation between ethanol treatment time and severity of uterine damage and kidney dysfunction, particularly in the EtOH-40 s group. Overall, the study successfully established a mouse model of endometrial injury using a minimally invasive transcervical ethanol perfusion technique, providing a basis for further research on IUA pathogenesis and therapies.

Summary: Multiple signaling pathways are involved in tumor development, making bispecific antibodies more suitable for treating complex tumor diseases. A novel PE-based bispecific immunotoxin designed in this study showed improved efficacy against HER2-positive tumors. Transcriptome sequencing and real-time PCR were used to verify the results, indicating the potential of these bispecific immunotoxins for anti-HER2 cancer therapeutics.

Extracellular vesicle-transmitted miR-671-5p alleviates lung inflammation and injury by regulating the AAK1/NF-KB axis

Summary: Menstrual blood-derived stem cells can alleviate acute lung inflammation and injury through their extracellular vesicle-transmitted miR-671-5p. This miRNA directly targets AAK1 and regulates the activation of NF-kB signaling. This study provides insights into the molecular basis of how extracellular vesicles from mesenchymal stem cells improve pulmonary inflammatory injury and highlights the functional importance of the miR-671-5p/AAK1 axis.

p38-mediated FOXN3 phosphorylation modulates lung inflammation and injury through the NF-κB signaling pathway

Summary: NF-kappa B activation leads to MRSA-induced lung inflammation, and FOXN3 can inhibit NF-kappa B signaling to reduce the inflammatory response. FOXN3 competes with I kappa B alpha for binding to hnRNPU, preventing I kappa B alpha degradation and NF-kappa B activation. Phosphorylation of FOXN3 by p38 induces dissociation from hnRNPU, leading to NF-kappa B activation. FOXN3 phosphorylation is clinically correlated with pulmonary inflammatory disorders.
