Therapy of lymphoma by immune checkpoint inhibitors: the role of T cells, NK cells and cytokine-induced tumor senescence

Fatima Ahmetlic, Josia Fauser, Tanja Riedel, Vera Bauer, Carolin Flessner, Nadine Hoemberg, Roman Hennel, Ellen Brenner, Kirsten Lauber, Martin Roecken, Ralph Mocikat

Summary: The study found that in treating lymphoma in mice with immune checkpoint inhibitors, 20% to 30% of mice were long-term protected and did not show clinical signs of tumor growth. The therapeutic effect depended on cytokine-induced senescence in malignant B cells. Therapy with immune checkpoint antibodies improved T cell functions and long-time survivors were only observed in the presence of T cells.

Case Report: Combined CDK4/6 and MEK Inhibition in Refractory CDKN2A and NRAS Mutant Melanoma

Andrea Forschner, Tobias Sinnberg, Gabi Mroz, Christopher Schroeder, Christian Philipp Reinert, Sergios Gatidis, Michael Bitzer, Thomas Eigentler, Claus Garbe, Heike Niessner, Martin Rocken, Cristiana Roggia, Sorin Armeanu-Ebinger, Olaf Riess, Sven Mattern, Dominik Nann, Irina Bonzheim

Summary: Limited treatment options exist for metastatic NRAS mutant melanoma patients resistant to immune checkpoint inhibitors. Molecular defects in cell cycle regulation can be additional challenges for these patients. Individualized therapy based on next-generation sequencing results can be effective, and further collaborative research is needed to expand treatment options for NRAS mutated melanoma.

Interleukin-10 counteracts T-helper type 1 responses in B-cell lymphoma and is a target for tumor immunotherapy

Yanchun Ma, Vera Bauer, Tanja Riedel, Fatima Ahmetlic, Nadine Homberg, Thomas P. Hofer, Martin Rocken, Ralph Mocikat

Summary: The study reveals that IL-10 suppresses ongoing immune responses in lymphoma and may be a potential target for immunotherapy.

Mesalazine Suppresses Proinflammatory Cytokines in Patients with Acute Anterior Uveitis Independently of HLA-B27

Nikola Smatlik, Nourhan Mortada, Martin Roecken, Amir S. Yazdi, Manfred Zierhut

Summary: The study revealed the inhibitory effect of 5-ASA on proinflammatory cytokines in PBMCs of patients with HLA-B27+ and HLA-B27- acute anterior uveitis. The results showed similarities between the two types of AAU but did not explain how HLA-B27 status affects the therapeutic response to 5-ASA.

Eigentler

Summary: Checkpoint inhibitors have greatly improved the treatment of metastatic melanoma patients. This study developed a novel method to estimate pretreatment metastatic growth rate (MGR) and found that high MGR is strongly associated with unfavorable survival outcomes in melanoma patients receiving anti-PD-1 antibodies.

Immune checkpoint blockade impairs immunosuppressive mechanisms of regulatory T cells in B-cell lymphoma

Vera Bauer, Fatima Ahmetlic, Nadine Hoemberg, Albert Geishauser, Martin Roecken, Ralph Mocikat

Summary: Research has shown that in malignant diseases, Tregs mediate immunosuppression through direct contact and IL-10. ICB antibodies may reduce Tregs' suppressive capacity by altering the inflammatory environment, contributing to the interference with Treg-dependent immunosuppression.

ASC Speck Formation after Inflammasome Activation in Primary Human Keratinocytes

Nikola Smatlik, Stefan Karl Drexler, Marc Burian, Martin Roecken, Amir Sadegh Yazdi

Summary: Chronic UV irradiation leads to various skin changes, including inflammation and alterations in pigmentation. UVB exposure induces increased NF-kappa B activation and the induction of inflammatory cytokines, playing a significant role in cell death and inflammation.

Low-dose total body irradiation facilitates antitumoral Th1 immune responses

Dominik Sonanini, Christoph M. Griessinger, Barbara F. Schoerg, Philipp Knopf, Klaus Dittmann, Martin Roecken, Bernd J. Pichler, Manfred Kneilling

Summary: The study demonstrated that low-dose TBI can significantly improve the efficacy of CD4+ T cell-based immunotherapies, leading to increased survival of RIP1-Tag2 mice and enhanced infiltration of CD4+ T cells in tumor tissue. This approach may provide a rationale for combining TBI with immunotherapy for cancer treatment in clinical settings.

Immunomodulatory role of reactive oxygen species and nitrogen species during T cell-driven neutrophil-enriched acute and chronic cutaneous delayed-type hypersensitivity reactions

Brandes, Martin Schaller, Wolfram Ruf, Martin Eichner, Kamran Ghoreschi, Martin Roecken, Bernd J. Pichler, Manfred Kneilling

Summary: ROS/RNS play a dominant role in acute and chronic delayed-type hypersensitivity reactions, with MPO and NOX2 identified as the main sources. However, depletion of only one primary source has marginal and conflicting effects on the outcomes of acute and chronic diseases, demonstrating that ROS/RNS are not a singular entity with different properties at various disease stages.

Prognostic role of gamma-glutamyl transferase in metastatic melanoma patients treated with immune checkpoint inhibitors

Johanna Winter, Max M. Lenders, Maximilian Gassenmaier, Andrea Forschner, Ulrike Leiter, Benjamin Weide, Mette-Triin Purde, Lukas Flatz, Antonio Cozzio, Martin Rocken, Claus Garbe, Thomas K. Eigentler, Nikolaus B. Wagner

Summary: The study found that the frequency of hepatic immune-related adverse events is underestimated, and the addition of gamma-glutamyl transferase (GGT) to laboratory testing can help detect hepatobiliary toxicity in patients receiving immune checkpoint inhibitors. Immune-related GGT elevations are associated with treatment response and survival outcomes.

The role of dendritic cells for therapy of B-cell lymphoma with immune checkpoint inhibitors

Anne Scheuerpflug, Fatima Ahmetlic, Vera Bauer, Tanja Riedel, Martin Roecken, Ralph Mocikat

Summary: The study demonstrates that immune checkpoint blocking therapy may involve dendritic cells in mediating anti-tumor responses through enhancing Th1-type immune response, in addition to directly activating T cells.
