Summary: Research has found that stress leads to upregulation of Fkbp5 mRNA in male and female mice, and pretreatment with a selective FKBP51 inhibitor can reduce stress-induced anhedonia. This suggests that FKBP51 may play an important role in integrating circulating stress hormones and serotonergic regulation of stress responses.

Summary: Skeletal muscle size can be remodeled through various cues, with Myod1, GR, and Nrf1 identified as key players in muscle-specific enhancer-promoter communication. The study reveals that these factors control gene expression through direct contacts, transcription factor Nrf1 regulation, and Ctcf-anchored chromatin loops, orchestrating myofiber size regulation in a myofiber-specific manner. Additionally, GR negatively regulates muscle mass and strength in mice by down-regulating anabolic pathways.

Summary: Liver health is crucial for overall human health, and the aryl hydrocarbon receptor (AHR) has been shown to play a significant role in liver diseases. Activation of the AHR pathway by gut microbiota may present therapeutic opportunities for liver diseases. By focusing on AHR signaling, a promising target in the gut microbiota can be identified for better understanding and treatment of liver diseases.

Summary: This study identified a novel molecular mechanism of FKBP5 in ALD, regulating alcohol-induced liver injury through the Hippo pathway and CXCL1 signaling. Loss of FKBP5 may serve as a potential target for the treatment of ALD.

Summary: This study found tissue-specific expression changes of receptor-regulatory factors in critically ill patients, which adaptively guide the action of glucocorticoids in different tissues. These findings are important for understanding the importance of glucocorticoid resistance and glucocorticoid treatment.

Summary: This study aimed to investigate the therapeutic effect of selective glucocorticoid receptor modulation with relacorilant in platinum-resistant/refractory ovarian cancer. The results showed that intermittent relacorilant plus nab-paclitaxel significantly improved progression-free survival, duration of response, and overall survival.

Summary: Efficient delivery of miR-122 to hepatocytes using a nanomedicine-based formulation has the potential to prevent and treat colorectal cancer liver metastasis. Mechanistically, it downregulates key genes involved in metastatic and cancer inflammation pathways, increases CD8(+)/CD4(+) T-cell ratio, and decreases immunosuppressive cell infiltration, making the liver more conducive to antitumor immune response.

Summary: Glucocorticoids are potent drugs with anti-inflammatory and immunosuppressive effects, but their clinical use is limited due to severe side effects. The molecular mechanisms underlying glucocorticoid-induced bone loss are unclear. In this study, RNA sequencing analysis identified Xist as a glucocorticoid-regulated gene involved in glucocorticoid-induced osteoporosis.

Summary: The impact of EphA2 receptor on cancer malignancy is determined by two different activation methods. PIP2 signaling lipid promotes TM dimerization in a conformation resembling the ligand-independent state, potentially affecting EphA2 activation. The difference in TM crossing angles in the ligand-dependent and ligand-independent states may play a role in regulating EphA2 activity.

Summary: Glucocorticoids (GCs) are steroid hormones that regulate various homeostatic functions in the body through the binding of glucocorticoid receptor (GR). GR signals are biased, with variations in different tissue cells, disease states, and ligands. This bias regulation of GR signaling depends on ligand-induced metameric regulation, protein post-translational modifications, assembly at response elements, context-specific assembly, and intercellular differences. Selective GR agonists and modulators (SEGRAMs) have been developed to bias therapeutic outcomes towards expected effects by influencing GR-mediated gene expression. This paper reviews the bias regulation and mechanism of GR, as well as the progress in drug research.

Summary: Estrogen plays a protective role against liver cancer in females primarily through Estrogen Receptor-alpha rather than Estrogen Receptor-beta. Deficiency of Estrogen Receptor-alpha significantly increases the risk of hepatocellular carcinoma in females, while deletion specifically in hepatocytes maintains female-specific liver gene expression. The control of sex-specific liver gene expression by Esr1, which mediates liver cancer risk, involves cells other than hepatocytes.
