The Flow Cytometry Standard (FCS) format is a widely accepted norm for storing Flow Cytometry (FCM) data. Its goal as a standard is to allow FCM data sharing and re-analysis. Over more than three decades of its existence FCS has evolved into a well-defined, flexible file format reflecting technical changes in the FCM field. Its flexibility as well as rising numbers of instrument vendors leads to suboptimal implementations of FCS in some cases. Such situations compromise the primary goal of the standard and hinder the ability to reproduce FCM analyses. It is further underlined by rapid rise of advanced FCM analyses, often carried out outside traditional software tools and heavily relying on standard data storage and presentation. We have developed flowIO, an R package which tests FCS file conformance with the standard as defined by International Society for Advancement of Cytometry (ISAC) normative. Along with the package we provide a web based application (also at ) allowing user friendly access to the conformance testing as well as FCS file editing and export for further analysis.

Summary: The study documented a BCP-ALL subtype with early switch to monocytic lineage and loss of B-cell immunophenotype, correlated with three genetic subtypes. The monocytic switch had significant impact on residual disease assessment and treatment outcomes.

Nucleoporin TPR Affects C2C12 Myogenic Differentiation via Regulation of Myh4 Expression

Jana Uhlirova, Lenka Sebestova, Karel Fiser, Tomas Sieger, Jindriska Fiserova, Pavel Hozak

Summary: The nucleoporin TPR plays a crucial role in the regulation of genes related to muscle formation and function and has been shown to affect the morphology of myoblasts and myotubes. However, more evidence is needed to fully understand how TPR specifically influences the expression of myogenic genes.

A novel class of ZNF384 aberrations in acute leukemia

Summary: A novel class of genetic aberration affecting the ZNF384 gene has been identified in leukemia cases, mimicking the phenotype of those with canonical ZNF384 fusions. This finding suggests that some ZNF384r-like cases represent the same genetic subtype as leukemia with canonical ZNF384 fusions.

Summary: This study collected data from 213 COVID-19 patients admitted to ten hospitals in Czech Republic, revealing weak evidence for the potential efficacy of azithromycin and favipiravir treatment. Additionally, the external validation of several proposed prognostic models for Covid-19 severity showed unsatisfactory performance on the dataset.
