Coulter volume is far more accurate measure of cell volume than forward angle light scatter. In this report, we have used Coulter volume to determine the mean cell volume and diameter of normal human peripheral blood cells and hematopoietic progenitor cells obtained by apheresis (HPC-A) from patients with hematological malignancies. Fresh peripheral blood samples (treated with Beckman Coulter IMMUNOPrep erythrocyte lysis solution), HPC-A samples (treated with BD Biosciences FACSLysing solution), or processed by Ficoll Hypaque sedimentation method were stained with CD45-FITC and PE-labeled CD34, CD90, CD117, and CD133 antibodies and analyzed for electronic volume and two color fluorescence. The mean electronic volume and diameter of mononuclear cells from fresh peripheral blood samples prepared with IMMUNOPrep were lymphocytes (191 mu m(3), 7.16 mu m), monocytes (370 mu m(3), 9.91 mu m), and granulocytes (328 mu m(3), 8.56 mu m). In mononuclear cells of HPC-A samples prepared by His-3 topaque-1077 sedimentation, the lymphocytes had volume and diameter of 311 mu m(3), 8.4 mu m, monocytes were 486 mu m(3), 9.76 mu m, and granulocytes were 515 mu m(3), 9.95 mu m. In contrast, HPC-A samples prepared after lysis with FACSLysing solution had mean electronic volume and diameter of lymphocytes (414 mu m(3), 9.25 mu m), monocytes (797 mu m(3), 11.5 mu m), and granulocytes (670 mu m(3), 10.85 mu m). Cell volume of mononuclear cells in the HPC-A samples prepared by Histopaque-1077 sedimentation method was correlated with the expression of stem cell markers CD34, CD90, CD117, and CD133. CD90 positive cells had the smallest mean electronic volume of 299.93 mu m(3)), when compared with cells with positive expression of CD133 (322 mu m(3)), CD 117 (349 mu m(3)), CD34 (407 mu m(3)), and CD45 (453 mu m(3)). Correlation of cell volume with stem cell marker expression may allow for the identification of small stem cells, which may not express the conventional markers used for the identification of stem cells in HPC-A samples. Correlation of Serum Selenium in Asthma Patients with Severity of the Disorder

Nidhi Girdhar, Heena Kansal, Kranti Garg, Siddharth Sharma, K. Sandeep Prabhu, Vishal Chopra, Alexey A. Tinkov, Anatoly Skalny, N. Tejo Prakash

Summary: This study investigated the correlation between serum selenium levels and the severity of asthma. The results showed that asthma patients had lower serum selenium levels compared to healthy individuals, with no significant differences based on gender and age. Patients with a family history of asthma and those using inhalers had slightly lower selenium concentrations.

Summary: This study aimed to identify single nucleotide polymorphisms (SNPs) of the MSH2 gene associated with cancer development using computational tools. Twenty-seven SNPs and five double amino acid change SNPs were found to potentially affect the structure and function of the MSH2 protein. Additionally, twelve identified SNPs were linked to specific types of cancer, such as hereditary nonpolyposis, colorectal cancer, and Lynch syndrome.

Immunotherapy in Small Cell Lung Cancer Treatment: a Promising Headway for Future Perspective

Harleen Kaur Walia, Parul Sharma, Navneet Singh, Siddharth Sharma

Summary: Despite being in the nascent stage, both prognosis and treatment for SCLC patients have made advancements with the introduction of immunotherapy as the fourth pillar of treatment. T cell checkpoint inhibitors show potential in anti-cancer activity and prolonging survival in SCLC patients by enhancing immune response. Immunotherapeutic approaches, such as TLR9 agonist and anti-Notch signaling, have also shown promise in clinical trials for SCLC.

Predictive role of polymorphic variants of phase II drug metabolising enzyme in modulating toxicity in North Indian lung cancer patients undergoing chemotherapy

Harleen Walia, Parul Sharma, Navneet Singh, Siddharth Sharma

Summary: This study investigated the associations between SNPs of Phase II detoxifying genes and platinum-induced toxicity in lung cancer patients. The findings showed that the NQO1 609 C > T polymorphism was associated with reduced risk of constipation and anorexia. The SULT1A1 Arg(213)His polymorphism was not associated with increased risk of nephrotoxicity. The NAT2 polymorphism exhibited increased risk of hematological toxicity.

Summary: This study used computational tools to predict the association between ABCC2 gene variants and Dubin-Johnson syndrome (DJS). The study found that 41 ABCC2 gene variants may be important etiological candidates for DJS in humans, and these variants had damaging effects on the function of the MRP2 protein, potentially leading to deficient bilirubin transportation capacity.

miRNAs are now starring in No Time to Die: Overcoming the chemoresistance in cancer

Anmol Bhatia, Atul Kumar Upadhyay, Siddharth Sharma

Summary: This article introduces the role of noncoding RNAs, such as miRNAs, in cancer treatment, as well as the research progress in combining miRNAs with anticancer drugs. It has been found that miRNAs play critical regulatory roles in key signaling cascades related to cancer cell proliferation, apoptosis, migration, and invasion. Combining miRNAs with anticancer drugs can effectively treat various types of cancer and improve the efficacy of treatment.

Genetic polymorphisms in the mismatch repair pathway (MMR) genes contribute to hematological and gastrointestinal toxicity in North Indian lung cancer patients treated with platinum-based chemotherapy

Sidhartha Singh, Navneet Singh, Siddharth Sharma

Summary: The present study found a relationship between polymorphisms in MLH1, MSH2, MSH3, and MSH6 genes and toxicity in North Indian lung cancer patients receiving platinum-based doublet chemotherapy. Certain gene polymorphisms were associated with an increased risk of anemia, leukopenia, neutropenia, and gastrointestinal toxicity. These polymorphisms could potentially serve as predictors for pretreatment evaluation in lung cancer patients.
