Summary: Research on bioactive components of herbal medicines has traditionally focused on secondary metabolites of herbal plants, with limited information on primary metabolites. This study highlights the immunostimulatory effects of heat-resistant RNA in a decoction of Glycyrrhizae Radix, suggesting nucleic acids as a new research field in the bioactive components of herbal medicines. By activating specific pathways and inducing cytokine production, the RNA demonstrated potential for therapeutic applications.

Summary: This study investigated the association of HIF-1 alpha expression and related mechanisms with latent or active tuberculosis infection. The results showed that IL-15 expression was higher in individuals with LTBI, while IL-18 and TNF-alpha expression were similar between LTBI and TB groups. Additionally, patients with active TB had higher serum 25(OH)D3 levels and expression of IL-6, HIF1A, and A20. The study also found that PBMCs from individuals with LTBI showed decreased NF-kappa B phosphorylation and increased TNF-alpha production after HIF-1 alpha blockade.

Summary: Long-term exposure to respirable silica particles can cause pulmonary inflammation and fibrosis, primarily promoted by cytokines released from alveolar macrophages. This study explored the mechanism of IL-1 beta biosynthesis and release from the perspective of nuclear factor kappa B (NE-kappa B). The results showed that silica nanoparticles (SiNPs) were internalized by macrophages, leading to cell damage. SiNPs exposure enhanced the expression of IL-1 beta and NE-kappa B, but this effect could be suppressed by BAY 11-7082, an NF-kappa B specific inhibitor. Animal experiments confirmed that silica exposure caused pulmonary inflammation and fibrosis, along with pathological changes in lung tissue.

Summary: Failure of bone healing after fracture often results in nonunion, but the underlying mechanism of nonunion pathogenesis is poorly understood. This study reveals that the inflammatory microenvironment in atrophic nonunion inhibits the expression of DNA methyltransferases, resulting in overexpression of CtBP1/2. The increased CtBP1/2 acts as transcriptional corepressors that suppress the expression of six genes involved in bone healing. The disruption of this signaling may represent a new therapeutic strategy to prevent nonunion incidence after bone fracture.
