GEF-H1/RhoA signalling pathway mediates lipopolysaccharide-induced intercellular adhesion molecular-1 expression in endothelial cells via activation of p38 and NF-κB

The purpose of study is to investigate the effects of GEF-H1/RhoA pathway in regulating intercellular adhesion molecule-1 (ICAM-1) expression in lipopolysaccharide (LPS)-activated endothelial cells. Exposure of human umbilical vein endothelial cells (HUVECs) to LPS induced GEF-H1 and ICAM-1 expression in dose-and time-dependent up-regulating manners. Pretreatment with Clostridium difficile toxin B-10463 (TcdB-10463), an inhibitor of Rho activity, reduced LPS-related phosphorylation of p65 at Ser 536 in a dose-dependent manner. Inhibition of TLR4 expression significantly blocked LPS-induced RhoA activity, NF-kappa B transactivation, GEF-H1 and ICAM-1 expression. Coimmunoprecipitation assay indicated that LPS-activated TLR4 and GEF-H1 formed a signalling complex, suggesting that LPS, acting through TLR4, stimulates GEF-H1 expression and RhoA activity, and thereby induces NF-kappa B transactivation and ICAM-1 gene expression. However, GEF-H1/RhoA regulates LPS-induced NF-kappa B transactivation and ICAM-1 expression in a MyD88-independent pathway because inhibition of MyD88 expression could not block LPS-induced RhoA activity. Furthermore, pretreatment with Y-27632, an inhibitor of ROCK, significantly reduced LPS-induced p38, ERK1/2 and p65 phosphorylation, indicating that ROCK acts as an upstream effector of p38 and ERK1/2 to promote LPS-induced NF-kappa B transactivation and ICAM-1 expression. What is more, the p38 inhibitor (SB203580) but not ERK1/2 inhibitor (PD98059) blocked LPS-induce NF-kappa B transactivation and ICAM-1 expression, which demonstrates that RhoA mediates LPS-induced NF-kappa B transactivation and ICAM-1 expression dominantly through p38 but not ERK1/2 activation. In summary, our data suggest that LPS-induced ICAM-1 synthesis in HUVECs is regulated by GEF-H1/RhoA-dependent signaling pathway via activation of p38 and NF-kappa B. (C) 2011 Elsevier Ltd. Summary: The double perovskite multiferroic materials Bi2FeCrO6 show significant improvement in magneto-electric, photovoltaic, and photocatalytic performance compared to BiFeO3. By using density functional theory, a novel double perovskite multiferroic material In2FeX (X = V, Cr, Mn, Co, and Ni) O6 is designed to achieve superior performance in ferroelectricity, magnetism, photovoltaics, and photocatalysis. The calculation results indicate that In2FeXO6 (X = V or Cr) O-6 could be a metastable multiferroic material with high ferroelectric polarization strength, driven by the interaction between In-O ions. In2FeVO6 exhibits a suitable bandgap and high photoelectric conversion efficiency as an excellent ferroelectric photovoltaic material. It can also absorb visible light to drive water splitting for hydrogen production under tensile strain.

Pedicled Omental Transposition for Recurrent Skin Erosion Following Deep Brain Stimulation Without Hardware Removal

Summary: This study aimed to evaluate the efficacy and safety of pedicled omental transposition in the treatment of recurrent skin erosion following deep brain stimulation. The results showed that patients in the pedicled omental transposition group had no symptoms during follow-up, and had significantly lower recurrence and explanation rates. This new surgical method offers an effective and safe solution for saving deep brain stimulation systems in patients with skin erosions.

Dynamical and Structural Properties of Comb Long-Chain Branched Polymer in Shear Flow

Summary: In this study, the effect of arms and shear flow on the dynamical and structural properties of the comb long-chain branched (LCB) polymer with dense arms was investigated using hybrid multi-particle collision dynamics (MPCD) and molecular dynamics (MD) methods. The results show that the LCB polymer undergoes periodic stretched-folded-stretched state transition and rotation, with the arms playing a role in stabilizing the polymer's motion. The impact induced by random and fast motions of arms and backbone decreases as the shear rate increases. The length of the arms and the conformational distribution are found to be important factors affecting the rotation period of the LCB polymer. The structural properties analysis reveals that the changes in gyration of the LCB polymer are in accordance with its rolling behavior, and the resistance to configuration expansion against the imposed flow field is stronger for the comb polymer with shorter arms. Additionally, with increasing arm length, the comb polymer transitions from linear-polymer-like to capsule-like behavior in shear flow.

Single-polymer dynamics of starch-like branched ring polymers in steady shear flow

Summary: The stretching dynamics and dynamical behaviors of individual branched ring polymer (BRP) were studied in steady shear flow. The nonmonotonic increase of polymer size with increasing branch length was observed. A universal power law described the decrease of alignment angle during the first downward phase. Tumbling and tank-treading motions were reflected by periodic and non-periodic changes of the polymer's ring backbone.

Multidimensional analysis using low-dose computed tomography to evaluate the severity of Mycoplasma pneumoniae pneumonia in children

Summary: This study analyzed low-dose computed tomography findings to assess the severity and progression of childhood Mycoplasma pneumoniae infection. The results showed that lobar consolidation is a stable and reliable CT feature that can be used to evaluate the severity of Mycoplasma pneumoniae pneumonia (MPP) in children. The number of lobes of consolidation was positively correlated with fever duration and the length of hospital stay, indicating its potential for predicting disease progression. Therefore, low-dose computed tomography is recommended for children with severe MPP.

This study investigated the potential involvement of macrophage-derived exosomes in the pathogenesis of cryptococcosis. The study analyzed the changes of microRNAs in macrophage exosomes and explored the effects of highly expressed exo-miRNAs on human endothelial cell permeability, ROS accumulation, and pyroptosis in bronchial epithelioid cells. The results showed that exo-miR-4449 plays a crucial role in the pathogenesis of cryptococcosis and holds promise as a significant biomarker for treatment.

Inflammatory marker levels in children with tobacco smoke exposure

Summary: Failure of bone healing after fracture often results in nonunion, but the underlying mechanism of nonunion pathogenesis is poorly understood. This study reveals that the inflammatory microenvironment in atrophic nonunion inhibits the expression of DNA methyltransferases, resulting in overexpression of CtBP1/2. The increased CtBP1/2 acts as transcriptional corepressors that suppress the expression of six genes involved in bone healing. The disruption of this signaling may represent a new therapeutic strategy to prevent nonunion incidence after bone fracture.
