Summary: This study validated a new method using ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) to quantify 12 epimers of ergot alkaloids in wheat. The method showed low LOD and LOQ values, allowing for sensitive detection of ergot epimers. The study also found differences between the R and S-epimers, which has rarely been documented in previous research.

Summary: The study aimed to develop the first validated method for analyzing human insulin and its synthetic analogues, primarily for plasma and post-mortem sera. Different extraction processes were evaluated for the lowest limit of detection for target analogues, and the limits of quantification were validated in plasma and post-mortem sera. Results showed that at the LOQ level, accuracy was between 70 and 130%, with precision lower than 30%.

Summary: A reliable and specific LC-MS/MS method was developed for determining ribociclib in both DBS samples and plasma. The method met FDA guidelines for validation and showed a strong correlation between ribociclib concentrations in DBS and plasma. This study demonstrates the potential of using DBS for monitoring ribociclib levels in breast cancer patients.

Summary: Standardization is crucial in lipidomics, with proposed traceable lipid class quantification for validation of workflows. Utilizing P-31 NMR and ICP-MS to validate LC-MS-based approaches, a novel quantification strategy based on lipid class separation and mass spectrometry is introduced. The concept proves interesting with low limits of detection and independent calibration strategies for quantitative assessment of lipid species.

Summary: Milk is a rich source of lipids, and LC-MS is the predominant technique for lipid identification and quantification. However, no standard methods are currently available for the quantification of milk phospholipids. This study aims to examine the factors influencing the quantification outcome of bovine milk phospholipids and discuss the pros and cons of current LC-MS methods.

Summary: To apply metabo-lipidomics in ophthalmological and systemic diseases, it is necessary to standardize the preanalytical and analytical steps in human tears. This study compared different methods for each step of the workflow and found that methanol/water extraction provided the best results for metabolomics, while Folch extraction was superior for lipidomics. The use of microcapillary glass tube for sampling was superior to Schirmer strip for metabolomics. A volume of 5 mu L provided satisfactory metabo-lipidomic coverage. These findings will contribute to improving the metabo-lipidomic workflow in human tears for the identification of new biomarkers.

Summary: This study developed and validated a LC-MS/MS method to quantify five tryptophan metabolites and investigated their correlation with body mass index (BMI) in pregnant women. The results showed a negative correlation between BMI and the analytes TRP, 5-HTP, and 5-HIAA, and a positive correlation with KYN.

Summary: The paper proposes a novel method of deep eutectic solvent (DES)-assisted liquid-liquid extraction (LLE) coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) for extracting steroid hormones from serum. The method has good linear calibration ranges, high extraction recoveries, and low limits of quantification, making it a simple and environmentally-friendly method.

Summary: A simple, fast, and sensitive tandem mass spectrometry method was established to detect the concentration of regadenoson in human plasma, which was used to study the pharmacokinetic characteristics of regadenoson in healthy Chinese subjects. The method showed linear calibration curve, low precision, high accuracy, and no matrix effect for regadenoson or regadenoson-d3.

Summary: In this study, two chromatographic methods based on direct saponification and n-hexane extraction were validated for the quantification of cholesterol in meat samples. Both methods showed good linearity, recovery, and precision. The GC-MS method exhibited higher sensitivity and lower limits of detection and quantification, making it a preferred method for cholesterol determination in meat.

Summary: Recent decades have shown an increase in the prescription of antipsychotic drugs in the European population, despite the presence of additional side effects. A new method for quantifying 38 antipsychotic drugs in plasma has been developed, revealing common use of second generation drugs like quetiapine and olanzapine, as well as continued presence of first generation drug haloperidol.
