Age-related macular degeneration (AMD) is a multifactorial eye disease that is associated with aging, family history, smoking, obesity, cataract surgery, arteriosclerosis, hypertension, hypercholesterolemia and unhealthy diet. Gender has commonly been classified as a weak or inconsistent risk factor for AMD. This disease is characterized by degeneration of retinal pigment epithelial (RPE) cells, Bruch's membrane, and choriocapillaris, which secondarily lead to damage and death of photoreceptor cells and central visual loss. Pathogenesis of AMD involves constant oxidative stress, chronic inflammation, and increased accumulation of lipofuscin and drusen. Estrogen has both anti-oxidative and anti-inflammatory capacity and it regulates signaling pathways that are involved in the pathogenesis of AMD. In this review, we discuss potential cellular signaling targets of estrogen in retinal cells and AMD pathology.

Summary: RPE replacement is an effective modern option for saving vision in cases of retinal degeneration caused by dysfunction of the retinal pigment epithelium. Cultivating RPE cells on PDLLA nanofibrous membranes results in better maturation and long-term survival, making them a plausible source for replacing non-functioning RPEs during cell therapy.

Summary: Age-related macular degeneration (AMD) is a complex disease characterized by damage to the retina and is a major cause of vision loss in elderly people. This review explores the role of non-coding RNAs in regulating mitochondrial function and antioxidant stress response in AMD. These molecules have the potential to be used as diagnostic markers and therapeutic targets for AMD and other oxidative stress-related diseases.

Summary: Injury of the cornea is a complex biological process that can be facilitated by mesenchymal stromal cells (MSCs) and tissue equivalents. This study presents a new tissue-engineering strategy for corneal stroma regeneration using cellularized 3D bioprinted hydrogel constructs implanted into cultured porcine corneas. The experiment successfully combines laboratory-based methods with patient-care practice to produce a cell-laden tissue equivalent for corneal implantation.

Biomacromolecular Profile in Human Primary Retinal Pigment Epithelial Cells-A Study of Oxidative Stress and Autophagy by Synchrotron-Based FTIR Microspectroscopy

Summary: Synchrotron radiation-based Fourier Transform Infrared (SR-FTIR) microspectroscopy is a technique used for non-destructive and chemically sensitive detection of biomolecular changes in cells. This study investigated the effects of autophagy and oxidative stress on human primary retinal pigment epithelium (hRPEs), using SR-FTIR microspectroscopy to analyze the changes in nucleic acids, proteins, carbonyl groups, and lipids. The results can be used to study drug discovery targeting autophagy and oxidative stress in age-related macular degeneration (AMD).

Dysphotopsias or Unwanted Visual Phenomena after Cataract Surgery

Summary: Dysphotopsias, divided into positive and negative types, are common visual disturbances that happen after cataract surgery. Positive dysphotopsias include glare, light streaks, starbursts, light arcs, rings, haloes, or flashes of light. Negative dysphotopsias appear as an arc-shaped shadow or line in the visual field. While positive dysphotopsias occur in a higher percentage of patients, persistent symptoms are rare. Negative dysphotopsias have a lower incidence rate, but more cases experience symptoms that persist beyond one year postoperatively.

This article provides an overview of the physical and chemical properties of vitreous substitutes, their clinical applications, and surgical techniques. The future developments in vitreous substitutes are extensively discussed, with a focus on desired outcomes and biomaterials technology.

Ex Vivo Pig Eye Model for Studying Diffusion of Acylcarnitines with Different Physicochemical Properties

Summary: This study explored the diffusion capacities between the anterior and vitreous chambers in a novel ex vivo pig eye model using stable isotope-labeled acylcarnitines and mass spectrometry analysis. The results showed a distinct diffusion pattern of molecules with different molecular size and hydrophobicity within and between the two chambers. This model can be valuable for optimizing therapeutic molecule choices and designs for intravitreal, intracameral, and topical treatments in the future.

Automatic Detection of Microaneurysms in Fundus Images Using an Ensemble-Based Segmentation Method

Summary: This study presents a novel method for automatically detecting microaneurysms in color fundus images. The proposed method consists of three main steps: image breakdown into smaller patches, inference to segmentation models, and reconstruction of the predicted segmentation map. The proposed segmentation method utilizes an ensemble of three different deep networks, including U-Net, ResNet34-UNet, and UNet++. The performance evaluation is based on Dice score and IoU values, and the ensemble-based model achieves higher scores compared to other network architectures. The proposed ensemble-based model demonstrates high potential for the practical application of early-stage diabetic retinopathy detection in color fundus images.

Inflammation and Oxidative Stress Gene Variability in Retinal Detachment Patients with and without Proliferative Vitreoretinopathy

Summary: This study examined the relationship between specific genetic variations and the risk of developing proliferative vitreoretinopathy (PVR) after surgery. The study analyzed single nucleotide polymorphisms (SNPs) in genes related to inflammation and oxidative stress, which are associated with PVR pathways, in 192 patients with primary rhegmatogenous retinal detachment (RRD) who underwent 3-port pars plana vitrectomy (PPV). The study found that certain SNPs, SOD2 rs4880 and IL1B rs1071676, were significantly associated with PVR risk. Carriers of the IL1B rs1071676 GG allele appeared to have better postoperative visual acuity among patients without PVR. These findings suggest that genetic variations may play a role in the development of PVR after surgery.

A Novel Technique of Amniotic Membrane Preparation Mimicking Limbal Epithelial Crypts Enhances the Number of Progenitor Cells upon Expansion

Summary: The study aimed to investigate if a new technique of human amniotic membrane (HAM) preparation, which mimics the crypts in the limbus, enhances the number of progenitor cells cultured ex vivo. The results demonstrated that the crypt-like HAMs expressed higher levels of progenitor markers p63 alpha, SOX9, and proliferation marker Ki-67 compared to flat HAMs, but no difference was found for the quiescence marker CEBPD. This novel HAM preparation method increased the number of progenitor cells expanded in the crypt-like HAM compared to cultures on the conventional flat HAM.

Summary: A systematic review was conducted on the use of antiplatelets and/or anticoagulants in vitreoretinal surgery. The findings suggest that the benefits of using these medications seem to outweigh the disadvantages, which are mainly related to postoperative hemorrhagic complications.
