Characterization of a new rat model for chronic inflammatory demyelinating polyneuropathies

Our objective was to develop a chronic model of EAN which could be used as a tool to test treatment strategies for CIDP. Lewis rats injected with S-palmitoylated P0(180-199) peptide developed a chronic, sometimes relapsing-remitting type of disease. Our model fulfills electrophysiological criteria of demyelination with axonal degeneration, confirmed by immunohistopathology. The late phase of the chronic disease was characterized by accumulation of IL-17(+) cells and macrophages in sciatic nerves and by high serum IL-17 levels. In conclusion, we have developed a reliable and reproducible animal model resembling CIDP that can now be used for translational drug studies. (C) 2014 Elsevier B.V. All rights reserved.

Summary: Neuromyelitis optica spectrum disorder (NMOSD) and multiple sclerosis (MS) are autoimmune inflammatory and demyelinating diseases of the central nervous system. Differentiating between NMOSD and MS is crucial for the timely and appropriate treatment. In this study, infrared spectroscopy combined with machine learning successfully distinguished NMOSD and MS patients with 100% sensitivity and specificity. The method shows promise as a cost-effective and rapid diagnostic tool to help save valuable treatment time for patients.

Anti-CD20 immunotherapy in progressive multiple sclerosis: 2-year real-world follow-up of 108 patients

These recommendations cover various aspects including pregnancy planning, follow-up, delivery routes, anesthesia, prevention of relapses, breastfeeding, vaccinations, etc. These recommendations are expected to harmonize counseling and treatment practices, and provide better and individualized choices for individuals with MS.

Myelin in Alzheimer's disease: culprit or bystander?

Summary: Alzheimer's disease (AD) is a neurodegenerative disorder characterized by the accumulation of toxic A beta peptide oligomers, plaques, and tangles containing tau protein, resulting in neuronal and synaptic losses. Sporadic AD, the more common form, is believed to be caused by chronic neuroinflammation and mitochondriopathies, leading to the accumulation of free radicals. Mutations in DNA and unfolded proteins in aging brains contribute to chronic amyloidosis, which affects myelin sheath and axons and ultimately leads to cognitive deficits and dementia. This study explores the potential role of early myelin alterations in the development of neuronal alterations and symptom onset, suggesting that structural and metabolic disorders in myelination/demyelination processes may play a crucial role in certain pathophysiological and clinical forms of the disease.
