Towards 50 years of platelet function analyser (PFA) testing

Emmanuel J. Favaloro, Leonardo Pasalic, Giuseppe Lippi

Summary: The platelet function analyser (PFA) is a widely used instrument for screening platelet function. It has two models, the PFA-100 and the PFA-200, with similar outputs and normal reference ranges. The PFA-100 has been in use for nearly 50 years, while the PFA-200 is expected to replace it in the future. The test system is highly sensitive to von Willebrand disease and aspirin therapy, but only moderately sensitive to platelet function defects and deficiencies. Its clinical utility varies according to user experience, with some researchers using it to assess thrombosis risk or pre-operative bleeding risk.

D-dimer: old dogmas, new (COVID-19) tricks

Giuseppe Lippi, Francois Mullier, Emmanuel J. Favaloro

Summary: D-dimer measurement is important in the diagnosis, exclusion, and prognostication of venous thromboembolism and disseminated intravascular coagulation. Its clinical applications are expanding to include other diseases such as acute aortic dissection, acute intestinal ischemia, and cerebral venous thrombosis, as well as the management of COVID-19.

?-sitosterol ameliorates inflammation and Pseudomonas aeruginosa lung infection in a mouse model

Alice Rossi, Alessandra Bragonzi, Melessike Medede, Ida De Fino, Giuseppe Lippi, Marco Prosdocimi, Anna Tamanini, Giulio Cabrini, Maria Cristina Dechecchi

Summary: We have previously demonstrated that beta-sitosterol (BSS) inhibits the expression of IL-8 in CF bronchial epithelial cells exposed to P. aeruginosa. In this study, we show that BSS significantly reduces leukocyte recruitment and bacterial colonization in the airways of a mouse model with chronic lung infection. BSS treatment also decreases the expression of key cytokines involved in immune response in the lung tissue. Our findings suggest that BSS has the potential to be a new drug for targeting excessive neutrophil recruitment in chronically infected lungs and further investigations on its protective mechanisms are encouraged.

Immunogenicity and Safety of the BNT162b2 COVID-19 Vaccine in Patients with Cystic Fibrosis with or without Lung Transplantation

Francesca Lucca, Valentino Bezzerri, Elisa Danese, Debora Olioso, Denise Peserico, Christian Boni, Giulia Cucchetto, Martina Montagnana, Gloria Tridello, Ilaria Meneghelli, Mirco Ros, Giuseppe Lippi, Marco Cipolli

Summary: This study investigated the immunogenicity and safety of the BNT162b2 vaccine in patients with cystic fibrosis (pwCF) and found that pwCF exhibited a similar immune response to the vaccine as the general population, while lung transplant recipients (LTR) showed impaired immunogenicity, especially in patients undergoing immunosuppressive treatment. The study results highlight the importance of longitudinal assessment of the immune response to COVID-19 vaccination.

Summary: In this study, the clinical performance of the SNIBE Maglumi SARS-CoV-2 antigen fully-automated chemiluminescent immunoassay (MAG-CLIA SARS-CoV-2 Ag) was reported. The results showed satisfactory analytical performance of MAG-CLIA SARS-CoV-2 Ag, which could be used as a surrogate for molecular testing in identifying high viral load samples.

Long-term stability of pleural fluid carcinoembryonic antigen and its effect on the diagnostic accuracy for malignant pleural effusion

Dan-Ni Yang, Yan Niu, Jian-Xun Wen, Xu-Hui Wen, Ya-Fei Wang, Li Yan, Ting-Wang Jiang, Jin-Hong Huang, Hong Chen, Giuseppe Lippi, Wen-Qi Zheng, Zhi-De Hu

Summary: This study aimed to investigate the long-term stability of pleural fluid carcinoembryonic antigen (CEA) at -80 degrees C to -70 degrees C and analyze the effects of frozen storage on the diagnostic accuracy of CEA for malignant pleural effusion (MPE). The results showed that CEA remains stable when stored at -80 degrees C to -70 degrees C for 1-3 years, and frozen storage does not significantly affect the diagnostic accuracy of CEA for MPE.
