Summary: The development of various cells, including hematopoietic stem cells and germ cells, relies on the receptor tyrosine kinase KIT and its ligand stem cell factor (SCF). Somatic mutations in KIT lead to several types of human cancers. Cryo electron microscopy analysis reveals that oncogenic KIT mutants exhibit distinct structural arrangements compared to wild-type KIT. The D5 region of KIT is identified as a potential target for therapeutic intervention.

Summary: The Class III receptor tyrosine kinase Flt3 and its ligand, the Flt3-ligand, play important roles in regulating hematopoietic and lymphoid progenitor cell proliferation, differentiation, and survival. They have also been found to be crucial in peripheral B cell development and affinity maturation. This review provides a comprehensive understanding of the molecular and cellular regulation of Flt3/FL and the roles of Flt3 signaling in hematopoietic stem cell activation, lymphoid development, bone marrow B lymphopoiesis, and peripheral B cell development. The literature confirms the significance of Flt3 signaling in B cell development and function, but also highlights the need for further research on Flt3-dependent stage-specific gene regulatory circuits in steady-state B lymphopoiesis.
