Summary: The performance of the Roche Elecsys Vitamin D total III assay for measuring total 25(OH)D was evaluated in this study. The results showed good agreement with liquid chromatography tandem mass spectrometry, and the assay demonstrated a higher biotin interference threshold and a broader measuring range compared to the previous generation assay. However, there was still an issue with cross-reactivity towards 3-epi-25(OH)D-3 in high titer samples.

Summary: This randomized controlled trial investigated the effect of high-dose vitamin D-3 on total bone mineral density (TtBMD) in the radius and tibia. The study found that increasing vitamin D dose was associated with increased plasma levels of 25-(OH)D-3, 24,25-(OH)(2)D-3, and 1,24,25-(OH)(3)D-3, but not 1,25-(OH)(2)D-3. These vitamin D metabolites were associated with a decline in TtBMD. The findings suggest that vitamin D metabolites other than 1,25-(OH)(2)D-3 may play a role in bone loss.

Summary: A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for quantification of vitamins D3 and D2 in serum. Results from two pilot clinical trials showed that serum and subcutaneous adipose tissue vitamin D3 concentrations increased proportionally to dose with 3 months of daily supplementation.

In this study, a diarylethene-based photoswitchable compound was separated and characterized using high-performance liquid chromatography, and the isomers were purified for individual study. Supercritical fluid chromatography was explored as an alternative separation method, offering faster analysis times and lower solvent consumption compared to high-performance liquid chromatography. This technique could be upscaled for future purification of diarylethene isomeric compounds, providing a more environmentally-friendly approach.
