Summary: We report the development and structural characterization of three humanized BTLA agonist antibodies, 22B3, 25F7, and 23C8. The crystal structures of the antibody-BTLA complexes show distinct and non-overlapping epitopes of BTLA. Among them, 22B3 mimics HVEM binding to BTLA and exhibits the strongest agonistic activity in functional cell assays and in an imiquimod-induced mouse model of psoriasis.

Summary: This study found that immune checkpoints HVEM, PD-1, and TIM-3 expressed on CD3(+)CD8(+) cells were effective in killing tumor cells in mice treated with paclitaxel. The dual blockade of BTLA on B cells and PD-1 on cytotoxic T cells significantly improved the therapeutic efficacy of paclitaxel against intraperitoneally disseminated tumors. Therefore, dual inhibition of BTLA and PD-1 has clinical potential for enhancing the efficacy of paclitaxel in treating intraperitoneal tumors, including ovarian carcinomas.

The study reveals that eNAMPT controls T-cell response through the PD-L1/PD-1 axis, and neutralization of eNAMPT restores antitumoral immune responses by activating CD8+IFN gamma+GrzB+ T cells and reducing the immunosuppressive phenotype of T regulatory cells.

Mechanistic studies revealed the potential of iRGD-anti-CD3 in promoting T cell extravasation through a transport pathway called vesiculovacuolar organelles in the endothelial cytoplasm.

Summary: Significant progress has been made in understanding the phenotypes of circulating immune cell sub-populations in human type 1 diabetes, but much less is known about the equivalent populations that infiltrate the islets to cause beta-cell loss. The phenotype and role of B-lymphocytes in the pancreas remain enigmatic, especially in relation to CD8+ T-lymphocytes. Differences in the proportion of islet-associated B-lymphocytes between different age groups suggest two endotypes of the disease, with CD8+ T-lymphocytes forming the predominant population in all groups.

Summary: The study revealed reduced BTLA expression on CD27(-)IgD(+) naive B cells from SLE patients, associated with disease activity, and impaired function. BTLA engagement was found to control CpG/TLR9 activation limiting plasmablast and B cell memory induction, but these functions were compromised in SLE B cells. Inhibition of SYK mimicked the effects of BTLA activity in vitro, suggesting a potential therapeutic principle for SLE.
