Co-expression of MMP-14 and MMP-19 predicts poor survival in human glioma

Matrix metalloproteinase (MMP)-14 and MMP-19 have been demonstrated to play an important role in the development of human gliomas. However, their prognostic values are not clear. The aim of this study was to investigate whether co-expression of MMP-14 and MMP-19 has prognostic relevance in human gliomas. Immunohistochemistry and western blot were used to investigate the expression of MMP-14 and MMP-19 proteins in 128 patients with gliomas. The expression levels of MMP-14 and MMP-19 proteins in glioma tissues were both significantly higher (both P < 0.001) than those in non-neoplastic brain tissues according to the immunohistochemistry analysis, which was confirmed by the western blot analysis. Additionally, the overexpression of either MMP-14 or MMP-19 was significantly associated with the advanced WHO grade (both P = 0.02), the low Karnofsky performance score (KPS) (P = 0.008 and 0.01, respectively) and the poor overall survival (both P = 0.01). Moreover, the Multivariate Cox proportional-hazards regression analysis revealed that the increased expressions of MMP-14 and MMP-19 were both independent prognostic factors for poor overall survival (both P = 0.02). Furthermore, the co-expression of MMP-14 and MMP-19 was additively and more significantly (P = 0.006) associated with adverse prognosis in patients with gliomas than respective expression of MMP-14 and MMP-19. These findings indicated for the first time that the co-expression of MMP-14 and MMP-19 is significantly correlated with prognosis in glioma patients, suggesting that the co-expression of these proteins may be used as both an early diagnostic and independent prognostic marker.

Summary: This study reveals the important role of m6A modifications in GBM tumor microenvironment (TME) cell infiltration and stemness characteristics. The m6A modification patterns are correlated with TME cell infiltration characteristics, stemness characteristics, tumor clinical outcome, immune infiltration, and stemness. Assessing the m6A modification pattern of individual tumors can enhance our understanding of TME infiltration and stemness characteristics and contribute to the development of immunotherapeutic strategies.

Wnt signaling regulates MFSD2A-dependent drug delivery through endothelial transcytosis in glioma

Summary: Inhibiting the Wnt signaling pathway can increase Temozolomide (TMZ) delivery and sensitize glioma to chemotherapy. Transcriptome analysis revealed that Wnt signaling inhibition enhances vascular transcytosis by upregulating PLVAP and downregulating MFSD2A. Additionally, Wnt signaling inhibition also enhances caveolae-mediated transcytosis in tumor endothelial cells (ECs). These findings suggest that Wnt signaling regulates MFSD2A-dependent TMZ delivery through a caveolae-mediated pathway, highlighting Wnt signaling as a promising therapeutic target for GBM treatment.

Prognostic and predictive factors of secondary gliosarcoma: A single-institution series of 18 cases combined with 89 cases from literature

Summary: This study assessed the clinicopathological features and outcomes of secondary gliosarcomas (SGS) through retrospective analysis of data. The results showed that initial diagnosis of GBM was a poor prognostic factor for SGS. Patients with initial non-GBM diagnosis who underwent gross total resection and chemoradiation had better overall survival outcomes. Patients initially diagnosed with GBM who received both chemoradiotherapy and active therapy after disease progression to SGS had significantly longer overall survival.

Real-time imaging of traumatic brain injury using magnetic induction tomography

Summary: In this study, an improved magnetic induction tomography (MIT) data acquisition system was used to monitor traumatic brain injury (TBI) and distinguish different injury levels. The experiment used a pneumatically controlled cortical impactor to strike the parietal lobe of anesthetized rabbits and establish different TBI models. The MIT data acquisition system continuously monitored the brain for an hour and successfully observed targets with increased conductivity in the reconstructed images, indicating that MIT may facilitate early TBI monitoring and distinguish varying degrees of injuries.

Summary: Hypoxia plays a critical role in glioblastoma by regulating the hypoxia-regulated genes (HRGs) network. CEBPD is identified as a key transcription factor that regulates a large number of hypoxia-regulated proteins (HRPs) and HRGs. CEBPD is up-regulated in glioblastoma and high levels are associated with poor prognosis. CEBPD activates the EGFR/PI3K pathway through ECM, particularly FN1-mediated EGFR phosphorylation.

Early detection of acute ischemic stroke using Contrast-enhanced electrical impedance tomography perfusion

Summary: A cerebral contrast-enhanced electrical impedance tomography perfusion method was developed for the early detection of acute ischemic stroke during thrombolytic therapy. The method utilized stable and highly conductive contrast agents and showed significant improvement in detection ability compared to other agents tested. This method accurately detected the location and area of cerebral infarction lesions and can be applied as a rapid, bedside imaging method for suspected ischemic stroke patients.

The Role of Hypoxia and Cancer Stem Cells in Development of Glioblastoma

Summary: Glioblastoma multiforme (GBM) is a highly malignant brain tumor characterized by a small population of glioblastoma stem-like cells (GSCs) that possess self-renewal, proliferative, invasive, and tumorigenic capabilities, leading to radio- and chemoresistance. Hypoxia inducible factors (HIFs) play a crucial role in the maintenance and progression of GSCs. This review summarizes the roles of hypoxia-associated GSCs in GBM development, including hypoxia-induced signatures, genes, pathways, and metabolic alterations. The concept of the hypoxic peri-arteriolar niche of GSCs is discussed. Autophagy and potential causes of therapeutic resistance in GBM are explored, along with chemotherapeutic agents to enhance the effectiveness of chemo-, radio-, or immunotherapy. Hyperbaric oxygen therapy (HBOT) is proposed as an adjuvant therapy to reverse the hypoxic microenvironment in GBM. Overall, understanding the intricate interactions between hypoxia and GSCs can lead to the development of novel therapeutic strategies to improve GBM patient survival.

The clinical and neurocognitive functional changes with awake brain mapping for gliomas invading eloquent areas: Institutional experience and the utility of The Montreal Cognitive Assessment

Summary: This study retrospectively analyzed the experience of a tertiary neurosurgical center in China over eight years in treating glioma patients with awake brain mapping. The results showed that awake brain mapping is a feasible and safe method for reducing the risk of neurological deficits, increasing extent of resection (EOR), and extending survival time. The MoCA test results indicated that brain mapping plays a critical role in preserving neurocognitive function during tumor resection.
