Low- and high-grade bladder cancer appraisal via serum-based proteomics approach

Objective: To address the shortcomings of urine cytology and cystoscopy for screening and grading of urinary bladder cancer (BC) we applied a serum-based proteomics approach as a surrogate tactic for rapid BC probing. Methods: This study was performed on 90 sera samples comprising of low-grade (LG, n = 33) and high-grade (HG, n = 32) BC, and healthy controls (HC, n = 25). Two-dimensional gel electrophoresis (2DE) tactic was executed to describe serum proteome. MALDI-TOF-MS (MS) was used to identify the characteristics of aberrantly expressed proteins in 2DE and validated using Western blot (WB) and ELISA approach. Receiver operating characteristics (ROC) curve analysis was also performed to determine the clinical usefulness of these proteins to discriminate among LG, HG and HC cohorts. Results: This comprehensive approach of 2DE, MS, WB and ELISA reveals five differentially expressed proteins. Among them two biomarkers (S100A8 and S100A9) were able to accurately (ROC, 0.946) distinguish 81% of BC (LG + HG) cases compared to HC with highest sensitivity and specificity. With a comparable tactic, two biomarkers (S100A8 and S100A4) were able to precisely (ROC, 0.941) discriminate 92% of LG cases from HG with utmost sensitivity and specificity. Conclusions: Serum proteomics probing appears to be an encouraging and least-invasive tactic for screening and grading of BC. (C) 2014 Elsevier B.V. All rights reserved.

A Study of Maternal and Umbilical Cord Blood Lead Levels in Pregnant Women

Summary: This study investigated the blood lead levels in pregnant women and umbilical cord blood, finding elevated lead levels in both groups. There was a strong positive correlation between the maternal and umbilical cord blood lead levels. Factors such as recent home painting and close proximity to traffic congestion were significantly associated with higher maternal blood lead levels. Education, mother age, fuel, and water sources did not show a significant association. Screening of blood lead levels and stricter regulations on lead-based products are recommended to reduce the exposure to lead in high-risk women and the general population.

Assessment of endocrine disrupting chemicals in breast milk: Association with dietary habits and duration of lactation

Summary: The present study aimed to assess the safety of infants in North India in relation to the presence of persistent organochlorine pesticides (OCPs) in breast milk, which could transfer endocrine-disrupting chemicals to newborns. Samples of colostrum and breast milk were collected and analyzed for pesticide concentration. Results showed that OCPs concentration was higher in breast milk compared to colostrum, indicating an increase in pesticides contamination over lactation. The study also found that women consuming non-vegetarian food had higher exposure to OCPs, and factors such as mother's age, gestational age, and infant birth weight were associated with OCPs levels in colostrum and breast milk. The study highlights the need for regulation and further research on the relationship between pesticide residues in breast milk and maternal and child health.

Genetics of congenital heart disease

Further research should focus on identifying common molecular mechanisms underlying this co-occurrence and promoting the research and treatment of developmental disorders related to the cardiovascular and central nervous systems.

Flow cytometry in the detection of circulating tumor cells in neoplastic effusions

Accurate identification of HLA-B*15:02 allele by two-dimensional polymerase chain reaction

Summary: This study successfully established a 2D-PCR method for identifying HLA-B*15:02 through a two-tube reaction. Among 1830 samples from the clinical general population, 3 HLA-B*15:02 homozygotes and 84 HLA-B*15:02 heterozygotes were detected.
