Summary: Reversed-phase HPLC is commonly used in MS-based proteomics for peptide separation, with particle-packed capillary columns being predominantly used in nanoflow HPLC systems. Despite capillary columns being a standard for many years, they are expensive and have short lifetimes, leading many laboratories to produce their own in-house packed columns.

Summary: Drugs of abuse testing is commonly used in clinical and forensic settings, with urine being the preferred specimen type but other emerging types include saliva, sweat, hair, and meconium. Liquid chromatography-tandem mass spectrometry (LC-tandem-MS) has become the new gold standard for confirmatory drug testing due to its ability to analyze a wider range of analytes compared to gas chromatography-mass spectrometry (GC-MS). Technical advancements such as high-resolution mass spectrometry and miniature ambient ionization mass spectrometry have improved the speed and efficiency of drugs of abuse testing.

Summary: Human breath contains trace amounts of non-volatile organic compounds (NOCs) which could be used as markers of individual health. In this study, metabolites and lipids in exhaled breath aerosol (EBA) samples were analyzed to identify molecular signatures of active tuberculosis (TB). Thirteen metabolite and nine lipid markers were identified with statistically different values between individuals with active TB and healthy controls. Importantly, EBA lipid profiles can effectively distinguish the two sample types, indicating the diagnostic potential of the identified molecules.

Summary: To apply metabo-lipidomics in ophthalmological and systemic diseases, it is necessary to standardize the preanalytical and analytical steps in human tears. This study compared different methods for each step of the workflow and found that methanol/water extraction provided the best results for metabolomics, while Folch extraction was superior for lipidomics. The use of microcapillary glass tube for sampling was superior to Schirmer strip for metabolomics. A volume of 5 mu L provided satisfactory metabo-lipidomic coverage. These findings will contribute to improving the metabo-lipidomic workflow in human tears for the identification of new biomarkers.

Summary: The cardiovascular system and the central nervous system exhibit a coordinated developmental process during embryonic development. Congenital heart disease (CHD) is the most common congenital disorder, and neurodevelopmental disorders (NDD) are common complications in CHD patients. Both genetic and non-genetic factors contribute to the co-occurrence of CHD and NDD. Further research should focus on identifying common molecular mechanisms underlying this co-occurrence and promoting the research and treatment of developmental disorders related to the cardiovascular and central nervous systems.
