Four newborn boxer dogs with bilateral cleft lip and palate were examined in search of further abnormalities, but no other type of congenital defect was found. These data, along with the pedigree inspection regarding the clinical phenotype, led to the conclusion that the affected dogs had a nonsyndromic monogenic autosomal recessive cleft lip and palate.

Summary: Through targeted next-generation sequencing of the EDA gene in a male poodle with X-linked hypohidrotic ectodermal dysplasia, it was found that the cause of the dysplasia is not point variants, small deletions, or insertions in the exons and splice sites of the gene. Patients with XLHED phenotype may have other types of variants in the EDA gene or in other genes of the EDA signaling pathway.

Summary: Inborn errors of metabolism are genetic disorders caused by a block in a metabolic pathway, affecting both humans and animals. They are individually rare diseases but relatively common as a group. Inbreeding in dog breeds increases the risk of disease recurrence due to the higher frequency of heterozygotes. Familiarity with this subject among general practitioners is significant in identifying new cases and increasing knowledge about inborn errors of metabolism.
