New Directions in Therapeutic Angiogenesis and Arteriogenesis in Peripheral Arterial Disease

Brian H. Annex, John P. Cooke

Summary: PAD is a significant global cause of disability and mortality, primarily due to atherosclerotic obstructions in the large arteries to the leg(s). Despite advances in surgical and endovascular procedures, treatment outcomes may be suboptimal at times due to comorbid conditions or severe microvascular disease.

Summary: This study aimed to determine the effects of prior COVID-19 on coronary microvasculature. By considering factors such as time from infection, disease severity, SARS-CoV-2 variants, and subgroups of patients with diabetes and those with no known coronary artery disease, the study compared the myocardial flow reserve (MFR) between patients with previous COVID-19 and controls. The results showed that patients with prior COVID-19 had a significantly higher risk of impaired MFR.

Telomerase mRNA Enhances Human Skin Engraftment for Wound Healing

David F. Chang, Karem A. Court, Rhonda Holgate, Elizabeth A. Davis, Katie A. Bush, Andrew P. Quick, Aldona J. Spiegel, Maham Rahimi, John P. Cooke, Biana Godin

Summary: Using autologous hSCS to treat deep skin wounds requires less donor skin and can prevent the decline of therapeutic effect caused by skin aging. Utilizing hTERT mRNA LNP can enhance the engraftment and proliferation of hSCS, reducing cellular senescence and DNA damage.

Mechanotransduction-on-chip: vessel-chip model of endothelial YAP mechanobiology reveals matrix stiffness impedes shear response

Brandon K. Walther, Navaneeth Krishna Rajeeva Pandian, Karli A. Gold, Ecem S. Kilic, Vineeth Sama, Jianhua Gu, Akhilesh K. Gaharwar, Anthony Guiseppi-Elie, John P. Cooke, Abhishek Jain

Summary: The study discusses the importance of endothelial mechanobiology in vascular dysfunction, focusing on the effects of shear stress and matrix stiffness on endothelial cell responses. By utilizing vessel-chip systems, researchers were able to model and validate the mechanobiological behaviors of endothelial cells in response to different mechanical stimuli, providing insights into the progression of diseases like atherosclerosis.
