The Tertiary Amine Nitrogen Atom of Piperazine Sulfonamides as a Novel Determinant of Potent and Selective β3-Adrenoceptor Agonists

Novel compounds were prepared in fair to good yields as human beta(3)-adrenoceptor (beta(3)-AR) agonists. In particular, aryloxypropanolamines 7a-d (EC50=0.57-2.1 nm) and arylethanolamines 12a,b,e (EC50=6.38-19.4 nm) were designed to explore the effects of modifications at the right-hand side of these molecules on their activity as beta(3)-AR agonists. Piperidine sulfonamides 15a-c,e-g (EC50=6.1-36.2 nm) and piperazine sulfonamide derivatives 20-29 (EC50=1.79-49.3 nm) were examined as compounds bearing a non-aromatic linker on the right-and left-hand sides of the molecules. Some piperazine sulfonamides were found to be potent and selective beta(3)-AR agonists, even if the amine nitrogen atom is tertiary and not secondary, as is the case for all beta(3)-AR agonists reported so far. (S)-3-{4-{N-{4-{2-[2-Hydroxy-3-(4-hydroxyphenoxy)propylamino]ethyl}phenyl}sulfamoyl}phenoxy}propanoic acid (7d; EC50=0.57 nM), (R)-N-{4-[2-(2-hydroxy-2-phenylethylamino)ethyl]phenyl}-4-(3-octyl-ureido)benzenesulfonamide (12e; EC50=6.38 nm), (R)-2-[1-(4-methoxyphenylsulfonyl)piperidin-4-ylamino]-1-phenylethanol (15f; EC50=6.1 nm), and (S)-4-{2-hydroxy-3-[4-(4-methoxyphenylsulfonyl)piperazin-1-yl]propoxy}pheno; (25; EC50=1.79 nm) were found to be the most potent beta(3)-AR agonists of the aryloxypropanolamine, arylethanolamine, piperidine sulfonamide, and piperazine sulfonamide classes, respectively. The two most potent compounds were identified as possible candidates for further development of beta(3)-AR agonists useful in the treatment of beta(3)-AR-mediated pathological conditions.

Summary: A sustainable and efficient two-step methodology for the synthesis of Thenfadil has been developed, utilizing a combination of reductive amination and Cu-catalyzed C-N coupling reactions in environmentally responsible deep eutectic solvents. Under optimized conditions, both reactions proceed smoothly with good yields and suppression of side products. A simplified workup procedure and the potential for scale-up and synthesis of analogs have also been demonstrated.

Summary: This study aimed to identify molecular biomarkers specific to ovarian serous adenocarcinoma (OSCA) for improved diagnosis. Genome-wide screening and validation analysis identified gene expression biomarkers that accurately discriminate OSCA tissues from healthy ovarian tissues and other tumor types. These biomarkers can potentially enhance the molecular diagnosis of OSCA and can be applied to liquid biopsies.

Olive Mill Wastewater Fermented with Microbial Pools as a New Potential Functional Beverage

Summary: The objective of this study was to develop a new functional beverage with health-promoting effects using olive mill wastewater (OMWW) as a raw material. OMWW was pre-treated and subjected to fermentation, resulting in a beverage with increased phenolic content and hydroxytyrosol. Fermentation also enhanced the antioxidant and anti-inflammatory activity of OMWW. This study confirmed that the process employed was an effective strategy to produce a new functional beverage.

Sigma-2 Receptor Ligand Binding Modulates Association between TSPO and TMEM97

Summary: Sigma-2 receptor (S2R) is functionally associated with both transmembrane protein TMEM97 and translocator protein (TSPO). This study provides evidence of the interaction between S2R and TSPO, as well as the physical colocalization of TMEM97 and TSPO in certain cancer cells. Additionally, the role of S2R-interacting partner PGRMC1 was clarified. This is the first suggestion of a functional interaction between TSPO and TMEM97, which can be influenced by S2R ligands.

Zoledronic Acid Blocks Overactive Kir6.1/SUR2-Dependent KATP Channels in Skeletal Muscle and Osteoblasts in a Murine Model of Cantu Syndrome

Summary: Cantu syndrome is caused by gain of function mutations in the ABCC9 and KCNJ8 genes, resulting in defects in the KATP channels. Glibenclamide has been proposed as a treatment, but it is not effective against certain mutations. Zoledronic acid was found to effectively reduce KATP currents in skeletal muscle and bone cells of Cantu syndrome mice, but not in SUR2 mutant mice. This indicates the importance of subunit specificity for appropriate therapy.
