A simple and efficient copper-catalyzed one-pot tandem method has been developed for synthesis of benzimidazo[1,2-b]isoquinolin-11-one derivatives via reactions of substituted 2-halo-N-(2-halophenyl)benzamides with alkyl 2-cyanoacetates or malononitrile under mild conditions.

Summary: The chiral phosphoric acid-catalyzed couplings of C-alkynyl N,N'-di-(tert-butoxycarbonyl)-aminals with beta-naphthols produced chiral propargylamines with moderate to high yields and high to excellent enantioselectivity. Sequential chiral phosphoric acid-catalyzed formation of N-(tert-butoxycarbonyl)-imines and 1,2-addition of beta-naphthols to the N-Boc-imines were key steps in the reactions. Additionally, chiral 1,2-dihydronaphtho[2,1-b]furans and naphtho[2,1-b]furans were successfully prepared by further adding AgOAc and 2,6-lutidine or 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) to the resulting chiral propargylamines solution.

Summary: A novel palladium-catalyzed [4 + 1] cycloaddition reaction has been developed for the synthesis of spirotetrahydrofuran oxindoles. This method utilizes diphenyl 2-oxoindolin-3-yl phosphates and 2-methylidenetrimethylene carbonate as reactants to provide the desired compounds in good to excellent yields at room temperature, offering a new strategy for the construction of spirooxindole derivatives with unique three-dimensional structures.

Electron Spin Catalysis with Graphene Belts

Summary: In this study, kinetic studies were conducted on spin catalysis reactions using electron spin resonance spectroscopy. Graphene belts synthesized by radical coupling method were used as catalysts. The results showed that sigma-type free radical species played a dominant role in the catalytic activity through spin-spin interaction. The spin catalysis mechanism was found to be applicable in both the oxygen reduction reaction and organic synthesis. The graphene belt spin catalyst demonstrated excellent performance with high ORR half-wave potential and long-term stability, as well as high conversion and yield in benzylamine coupling with molecular oxygen to generate imine.

Base-Controlled Palladium-Catalyzed Intramolecular 'One Substrate - Five Reactions' of 5-Benzyl-1-(2-halobenzyl)-2-phenyl-1H-pyrazol-3(2H)-ones

Summary: Achieving site-selectivity and chemoselectivity is challenging for substrates with multi-reactive sites in organic reactions. In this study, four different reactions were explored using model substrates with six reactive sites. By varying the reaction conditions, different bases were found to control the site-selectivity and chemoselectivity of the reactions. This strategy of "one substrate - multiple reactions - multiple products" reduces cost, increases product diversity, and provides more opportunities for pharmaceutical screening.

Pd/Brønsted acid catalysed intramolecular N-allylation of indoles and pyrroles with alkynes for the synthesis of N-fused heterocycles

Summary: In this article, a novel catalytic reaction using Pd(0) and Bronsted acid is reported for the synthesis of biologically important imidazolidinone-fused N-heterocycles through redox-neutral intramolecular N-allylation of indoles and pyrroles with alkynes. This atom-economical method is applicable to a wide range of substrates and eliminates the need for leaving groups or oxidizing agents commonly used in traditional allylation reactions.

Rh(III)-catalyzed selective mono- and dual-functionalization/cyclization of 1-aryl-5-aminopyrazoles with iodonium ylides

Modified minimal-size fragments of heparan sulfate as inhibitors of endosulfatase-2 (Sulf-2)

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Reductive cyclodimerization of chalcones: exploring the self-adaptability of galvanostatic electrosynthesis

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.

Rh(III)-catalyzed oxidative [4+2] annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols

Summary: In this study, the synthesis of functionalized benzo[a]phenazines and indazolo[2,3-a]quinolines was achieved through Rh(III)-catalyzed oxidative annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols. The method showed a wide substrate scope, excellent functional group tolerance, good to high yields of annulated products, and the capability for scaled-up synthesis. A tentative mechanism of the annulation reaction was proposed based on a preliminary mechanistic investigation.
