Viable route and DFT study for the synthesis of optically active limonaketone: A barely available natural feedstock in Cedrus atlantica

Summary: A novel, efficient synthesis pathway for optically active limonaketone was developed, starting from limonene, with an overall yield of 84%. The strategy involved a three-step total synthesis of limonaketone, a potentially important intermediate in limonene ozonolysis. By utilizing density functional theory calculations, the chemoselectivity in the Zn-deoxygenation reaction and its mechanistic pathway were elucidated.

Deconjugative α-Alkylation of Cyclohexenecarboxaldehydes: An Access to Diverse Terpenoids

Summary: This study presents a general and efficient method for the deconjugative alpha-alkylation of alpha,beta-unsaturated aldehydes under mild conditions through a synergistic effect between (BuOK)-Bu-t and NaH. The resulting beta,gamma-unsaturated aldehyde can be transformed into the corresponding allyl acetate via lead(IV) acetate-mediated oxidative fragmentation, with high yield. This strategy is applicable for constructing the carbon skeleton of diverse alkyl or aryl terpenoids.

Summary: A new strategy for the semisynthesis of the aromatic cassane-type diterpene taepeenin F (6) is reported, which showed potential antiproliferative activity against tumor cell lines and anti-inflammatory effects. Salicylaldehyde 21 was identified as the most active compound, inducing apoptosis in tumor cells and inhibiting inflammatory response.

Summary: Two main products, 18-hydroxy-cis-clerodan-3-ene-15-oic acid and 18-acetoxy-cis-clerodan-3-ene-15-oic acid, were isolated from the plant Cistus monspeliensis. The 18-acetoxy acid was converted to its mother 18-hydroxy acid in good yield. The synthesized 1,3,4-oxadiazole derivatives showed interesting antioxidant and antibacterial activities, as well as higher anti-inflammatory effects compared to the reference compound.

Summary: The acid treatment of 6,7-seco-abietane dialdehydes leads to the synthesis of derivatives with the 4a-methyltetrahydrofluorene skeleton of taiwaniaquinoids in high yield. The proposed mechanism involves the elimination of formic acid from the cyclic aldol intermediate. This novel reaction enables the enantiospecific synthesis of bioactive natural cupresol and taxodal.

Discovery of a synthetic taiwaniaquinoid with potent in vitro and in vivo antitumor activity against breast cancer cells

Summary: In this study, the antitumor activity of taiwaniaquinoids, specifically compound C36, was investigated in vitro and in vivo. Results showed that C36 exhibited significant antitumor effects on MCF-7 cells by inducing apoptosis through caspase 9 activation and an increase in the Bax/Bcl-2 ratio. In vivo experiments also demonstrated the efficacy of C36 in reducing tumor growth and improving survival rates in immune-competent C57BL/6 mice implanted with E0771 mouse mammary tumor cells.

Pd/Brønsted acid catalysed intramolecular N-allylation of indoles and pyrroles with alkynes for the synthesis of N-fused heterocycles

Summary: In this article, a novel catalytic reaction using Pd(0) and Bronsted acid is reported for the synthesis of biologically important imidazolidinone-fused N-heterocycles through redox-neutral intramolecular N-allylation of indoles and pyrroles with alkynes. This atom-economical method is applicable to a wide range of substrates and eliminates the need for leaving groups or oxidizing agents commonly used in traditional allylation reactions.

Modified minimal-size fragments of heparan sulfate as inhibitors of endosulfatase-2 (Sulf-2)

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Reductive cyclodimerization of chalcones: exploring the self-adaptability of galvanostatic electrosynthesis

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.

Rh(III)-catalyzed oxidative [4+2] annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols

Summary: In this study, the synthesis of functionalized benzo[a]phenazines and indazolo[2,3-a]quinolines was achieved through Rh(III)-catalyzed oxidative annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols. The method showed a wide substrate scope, excellent functional group tolerance, good to high yields of annulated products, and the capability for scaled-up synthesis. A tentative mechanism of the annulation reaction was proposed based on a preliminary mechanistic investigation.
