Summary: This study found that conditioned medium obtained from human tonsil-derived MSCs (T-MSC CM) can inhibit adipocyte differentiation by regulating glucocorticoid signal transduction, thereby reducing obesity. The experiments showed that T-MSC CM can attenuate early-stage adipocyte differentiation and protect mice from the negative effects induced by glucocorticoid treatment. Additionally, T-MSC CM enhances megakaryocyte differentiation and protects blood platelet counts.

Recent studies have revealed the molecular composition and clinical indications of EVs derived from adult mesenchymal stromal cells (MSCs). These EVs maintain some of the properties of their parent cells and have important implications for cell-free therapies.

Summary: Bone marrow adipose tissue (BMAT) is a significant component of the bone marrow space in healthy adults, but its expansion is associated with aging, obesity, anorexia nervosa, and irradiation. While traditionally viewed as a negative component, recent studies have revealed that BMAT can serve as an energy reservoir and also plays a role in regulating bone formation and hematopoiesis. This review summarizes the unique characteristics of BMAT and provides an updated understanding of its physiological roles based on a newly established mouse model.

Summary: Both adipose-derived and bone marrow-derived mesenchymal stem cells (ADSCs and BMSCs) can suppress renal fibrosis, with ADSCs showing a stronger effect under serum-containing culture conditions. However, ADSC infusion in mice may lead to a higher death rate from pulmonary embolism. Therefore, the selection of ADSCs or BMSCs for clinical applications should consider the target disease and culture method.

Summary: Cell-based therapies using PDLSC may be a promising alternative to MSC(M) and MSC(AT) for periodontal regeneration, as PDLSC showed enhanced proliferative and osteogenic potential. PDLSC expressed osteogenic markers and had higher calcium content and upregulated osteogenic/periodontal genes compared to MSC(M) and MSC(AT). CD146 was specifically expressed by PDLSC, which also had a higher proliferative capacity. However, alkaline phosphatase activity did not increase in PDLSC.

Summary: This study aimed to determine region-specific variations and age- and gender-specific differences in bone marrow adipose tissue (BMAT) in healthy subjects. The results showed distinct distribution of BMAT throughout the human skeleton, which was associated with age and gender. Furthermore, the study found that BMAT unsaturation was highest within the axial skeleton compared to the appendicular skeleton.

Results show that adipose tissue-derived MSCs from full term pregnant female sheep exhibit higher potential for proliferation and differentiation compared to male sheep-derived MSCs. Cryopreserved MSCs have similar characteristics to fresh cells. Therefore, it is important to consider tissue type and donor physiological status when applying MSCs in clinical settings.

Summary: BMAT is now recognized as a metabolically active organ that plays versatile roles in endocrine function, hematopoiesis, bone homeostasis, and metabolism. Age-related inflammatory changes associated with BMAT accrual and the downstream effect on endocrine function, energy expenditure, and metabolism are discussed, with potential therapeutic strategies addressed to prevent dysfunction during aging. Further research is needed to fully understand the explicit characterization of BMAT in aging.

Summary: This study analyzed the morphological and immunophenotypic changes of bone marrow adipocytes (BMAds) in different types of bone marrow cancers. The results showed that BMAds were significantly reduced in both bone marrow metastasis and myeloproliferative neoplasia with grade-3 myelofibrosis, along with a decrease in cell diameter and area. Additionally, the expression of certain markers in BMAds differed in these two types of cancers.
