Summary: RB and p53 are central tumor suppressors that play key roles in regulating the cell division cycle. RB downregulates numerous genes by forming complexes with E2F transcription factors, and its repression is released through phosphorylation. p53 activates CDKN1A gene to regulate RB phosphorylation, leading to expression of cell cycle regulators. There is an overlap between RB and DREAM signaling pathways.

Summary: Recent studies have shown that Haspin, a protein kinase essential for mitosis, plays a crucial role in the interphase progression of cancer cells. Loss of Haspin results in cell cycle defects, prolonging S/G2 phase, leading to mitotic cell death or postmitotic G1 arrest. Additionally, Haspin deficiency induces p21 checkpoint protein expression and co-depletion with p21 or p53 can partially restore cell proliferation.

Summary: Cell cycle regulators have been found to prevent cancerogenesis by acting as inhibitors or activators, and they also play active roles in differentiation, apoptosis, senescence, and other cellular processes. Recent evidence suggests that they are involved in bone healing/development cascade. Our study showed that deletion of p21, a cell cycle regulator, at the G1/S transition, enhanced bone repair capacity after a burr-hole injury in mice. Similarly, another study demonstrated that inhibition of p27 can increase bone mineral density and bone formation. In this review, we provide a concise overview of cell cycle regulators in osteoblasts, osteoclasts, and chondrocytes during bone development and/or healing. Understanding the regulatory processes of cell cycle during bone healing and development is crucial for the development of novel therapies to improve bone healing in aged or osteoporotic fractures.

Summary: ZNF674-AS1 expression is decreased in NSCLC compared to normal tissues, and its downregulation is significantly correlated with advanced TNM stage and decreased overall survival of NSCLC patients. ZNF674-AS1 inhibits NSCLC cell proliferation, colony formation, and tumorigenesis, accompanied by G0/G1 cell cycle arrest, through upregulation of p21 and downregulation of miR-423-3p. This study suggests the therapeutic potential of ZNF674-AS1 in NSCLC treatment.

Summary: This study evaluated the potential of four regeneration chemistries for desorbing phosphorus from a commercially available HIX-Nano resin hybridized with iron oxide nanoparticles. The combination of recovered NH4OH and tap water showed the highest phosphorus recovery potential, with low cost, high efficiency, and minimal impact on resin longevity. This technology may offer a cost-effective and sustainable solution to the global phosphorus problem in wastewater treatment applications.

Summary: FOXD1 is highly expressed in head and neck squamous cell carcinoma and over-expression is associated with poor prognosis. Knockdown of FOXD1 increases cellular senescence and reduces proliferation, while overexpression has the opposite effect. FOXD1 binds to the p21 promoter, inhibiting its transcription and blocking the CDK2/Rb signaling pathway, thereby preventing senescence and promoting tumor cell proliferation. miR-3oe-5p acts as a tumor suppressor by repressing FOXD1 translation, suggesting that FOXD1 may be a potential therapeutic target for HNSCC.

Summary: Xenin25 has an inhibitory effect on the spontaneous circular muscle contractions in the rat distal colon, with postinhibitory spontaneous contractions at a higher frequency. The mechanism involves activation of neuronal NTSR1, transmitters from VIP and apamin-sensitive NO neurons, and NK1R to modulate the muscle contractions. Xenin25 is considered an important modulator of postprandial circular muscle contraction in the distal colon.
