Fibulin-2 is involved in early extracellular matrix development of the outgrowing mouse mammary epithelium

Cell-matrix interactions control outgrowth of mammary epithelium during puberty and pregnancy. We demonstrate here that the glycoprotein fibulin-2 (FBLN2) is strongly associated with pubertal and early pregnant mouse mammary epithelial outgrowth. FBLN2 was specifically localized to the cap cells of the terminal end buds during puberty and to myoepithelial cells during very early pregnancy (days 2-3) even before morphological changes to the epithelium become microscopically visible, but was down-regulated thereafter. Exposure to exogenous oestrogen (E2) or E2 plus progesterone (P) increased Fbln2 mRNA expression in the pubertal gland, indicating hormonal control. FBLN2 was co-expressed and co-localised with the proteoglycan versican (VCAN) and co-localised with laminin (LN), while over-expression of FBLN2 in HC-11 cells increased cell adhesion to several extracellular matrix proteins including LN and fibronectin, but not collagens. Mammary glands from Fbln2 knockout mice showed no obvious phenotype but increased fibulin-1 (FBLN1) staining was detected, suggesting a compensatory mechanism by other fibulin family members. We hypothesise that similar to embryonic aortic smooth muscle development, FBLN2 and VCAN expression alters the cell-matrix interaction to allow mammary ductal outgrowth and development during puberty and to enable epithelial budding during pregnancy.

Summary: This study investigated the clinical features and genetic analysis of patients with tuberous sclerosis complex presenting with large rhabdomyoma tumors, as well as the potential role of autophagy and apoptosis in tumor pathogenesis. Results showed increased expression of autophagic and apoptotic markers in tumor cells compared to normal tissue, with patients carrying pathogenic mutations in the TSC-2 gene requiring urgent intervention. Future studies may lead to personalized treatment for cardiac rhabdomyoma.

Summary: Cardiac valves - highly complex structures that interact with cells, extracellular matrix, and hemodynamic environment - are tightly regulated by various mechanisms including epigenetic factors. This study analyzed the differential methylation profiles of non-diseased human aortic and mitral valve tissue, revealing candidate genes and signaling pathways associated with valve physiology and pathology. These findings contribute to our understanding of valve biology, tissue engineering approaches, and drug target discovery.

Systematic analysis of different degrees of haemolysis on miRNA levels in serum and serum-derived extracellular vesicles from dogs

Summary: This study investigated the effects of haemolysis on circulating miRNAs in dogs, and found that some miRNAs were significantly sensitive to haemolysis. The study also revealed differences in canine miRNA abundance compared to human miRNA. These findings have important implications for the discovery of circulating biomarkers in dogs, and emphasize the need to carefully consider the impact of haemolysis in serum or plasma-based tests.

Summary: We demonstrate a novel approach to generate atrial-like cardiomyocytes (CMs) from human-induced pluripotent stem cells (hiPSCs) through the treatment of Gremlin 2 and retinoic acid. The generated CMs exhibited similar morphology and characteristics to adult atrial myocytes (AMs), including expression of CM proteins and striated appearance. Single-cell RNA sequencing also revealed high expression of atrial-specific transcripts in the generated CMs. This method provides a valuable tool for studying atrial cell signaling pathways and atrial fibrillation (AF).

A matrisome RNA signature from early-pregnancy mouse mammary fibroblasts predicts distant metastasis-free breast cancer survival in humans

Summary: The study identified significant differences in extracellular matrix distribution in the mouse mammary gland during early pregnancy and found that pregnancy-associated fibroblasts enhanced branching initiation compared to control fibroblasts. A signature of differentially expressed matrisome RNAs was a strong prognostic indicator of distant metastasis-free survival in breast cancer patients, with particular relevance to estrogen receptor-negative breast cancers.
