Summary: This study successfully isolated and cultured cHDPCs and ncHDPCs from healthy and inflamed dental pulp tissue respectively. The growth characteristics and differentiation abilities were not significantly different between the two types of HDPCs. However, the expression of certain genes such as TLR-2 and IL-6 was significantly higher in cHDPCs, indicating an inflammatory phenotype in the inflamed dental pulp.

Summary: Periodontitis is a challenging inflammatory condition characterized by the deterioration of tooth support structures. Regeneration of periodontal tissues using stem cells, such as dental mesenchymal stem cells (DMSCs), non-dental stem cells, and induced pluripotent stem cells (IPSCs), shows promise but remains a challenge in clinical practice due to their complex physiological characteristics and the surrounding inflammatory microenvironment.

Summary: This study found that long non-coding RNA 00638 (lncRNA00638) regulates the osteogenic differentiation of periodontal ligament stem cells (PDLSCs) from periodontitis patients under static mechanical strain (SMS) loading by competing with miR-424-5p to target fibroblast growth factor receptor 1 (FGFR1). This finding may provide evidence for optimizing orthodontic treatments in patients with periodontitis.

Summary: The study reveals the crosstalk between dental pulp and dental follicle through the OGN-Hh pathway, which regulates the stemness and multidirectional differentiation of dental follicle stem cells. Ogn(-/-) mice exhibit accelerated root growth rate and thicker hard tissues in the root, suggesting the activation of the Hh pathway and increased cell proliferation and differentiation. This study provides important insights into the molecular regulation of early periodontal tissue development and lays a theoretical foundation for tooth root biology and periodontal tissue regeneration research.

Summary: The study revealed that periodontal ligament (PDL) cells exhibit dynamic proliferative characteristics during orthodontic tooth movement-induced tissue remodeling, with proliferative cells likely undergoing further division and contributing to tissue remodeling. Additionally, proliferating cells expressed various molecular markers, indicating the lineage plasticity of PDL cells in vivo.

Summary: The aim of this study was to evaluate the 20-year clinical outcomes of tissue-level implants in patients with a history of periodontitis and in periodontally healthy patients. The results showed that tissue-level implants, placed after comprehensive periodontal therapy and supportive periodontal care, had favorable long-term results. However, patients with a history of periodontitis who did not comply with supportive periodontal care were at higher risk of complications and implant loss.

Summary: Adult mesenchymal stem cells obtained from dental structures have the potential for complete regeneration and repair in various tissues, making them valuable for regenerative dentistry and medicine. Tissue engineering plays a crucial role in sustaining and enhancing the efficacy of these stem cells. Clinical trials are needed to ensure safety and improve the treatment outcomes for different pathologies. From a bioethical and anthropological perspective, the unique nature of the human individual facilitates personalized therapy, particularly for dental and chronic systemic diseases.

Summary: In a hypoxic microenvironment, ucMSCs enhance endothelial cell proliferation and migration through exosome-mediated paracrine function, accelerating wound healing. miR-125b in hypoxic exosomes targets and suppresses TP53INP1, alleviating hypoxia-induced cell apoptosis and promoting cutaneous repair.

Summary: This study comprehensively revealed the complexities of the molecular components in the osteoimmunology microenvironment of periodontal tissues at the single-cell level using scRNA-seq. Differences in specific cell types were observed in patients with periodontitis, with significant differentiation characteristics observed in certain cell types after initial periodontal therapy.

Summary: Current therapy for delayed replantation of avulsed teeth lacks predictability in promoting periodontal healing. This systematic review found that cell-based therapies significantly increased formation of new periodontal ligament and reduced adverse healing outcomes, highlighting their potential clinical benefits and translational value. Future research should focus on minimizing bias in animal experiments and standardizing outcome measures reporting to facilitate meta-analyses.

Summary: This study reveals the important role of cytokine receptor gamma c in thymic iT cell differentiation. It controls the cellular abundance, lineage commitment, and subset differentiation of iT cells. The abundance of gamma c proteins influences the generation of innate CD8 T cells and subset composition of invariant NKT cells in the thymus.

Summary: This study investigates the cytokine requirement for in vivo iNKT cells and finds that IL-7, not IL-15, is the homeostatic cytokine for iNKT cells. The abundance of IL-7 limits the size of the peripheral iNKT cell pool and there is competition for IL-7 between iNKT and conventional ab T cells.

Summary: Periodontitis is an oral disease caused by bacterial infection that leads to tissue destruction and tooth loss. When traditional treatment methods are not effective, tissue regeneration therapy using bone marrow-derived mesenchymal stem cells (BM-MSCs) and bone morphogenetic protein 7 (BMP7) can be considered. This study aimed to evaluate the efficacy of BMP7-expressing engineered BM-MSCs in promoting periodontal tissue regeneration in a rat model of periodontitis.

Summary: Innate CD8 T cells are terminally differentiated effector cells that resemble memory cells and exhibit proinflammatory properties. They differ from conventional effector-memory CD8 T cells in that their functional maturity is acquired during thymic development. This study identifies the cytokine receptor ?c as a critical mediator of innate CD8 T cell generation, promoting their selection even in the absence of classical MHC-I molecules. The expansion of IL-4-producing invariant NKT cells, driven by increased availability of intrathymic IL-4, plays a key role in the selection of innate CD8 T cells.
