Summary: Cell death plays a critical role in the pathogenesis of neurological diseases and brain cancers, with abnormal activation of PCD pathways being a common feature. Controlling PCD pathways may be a key component of future therapeutic strategies for these diseases.

Summary: BH3 mimetic drugs have the potential to trigger programmed cell death (PCD) and improve the treatment of glioblastoma multiforme (GBM). Co-targeting the pro-survival proteins BCL-XL and MCL-1 is more effective in killing GBM cells compared to conventional therapy, and combining with inducers of ferroptosis enhances the cell killing effect.

Summary: APR-246 can kill malignant cells irrespective of their TP53/TRP53 status. It induces various programmed cell death pathways, such as apoptosis, necroptosis, and ferroptosis. Therefore, the clinical testing of APR-246 should be expanded to cancers expressing wild-type TP53 or TP53-deficient.

Summary: This study demonstrates that megakaryocytes possess a functional necroptosis signaling cascade, but necroptosis in megakaryocytes is dispensable and does not impact platelet production. Platelet production was normal in the absence of MLKL, however, platelet activation and hemostasis were impaired with prolonged re-bleeding times.
