Co-culture of bone marrow-derived mesenchymal stem cells overexpressing lipocalin 2 with HK-2 and HEK293 cells protects the kidney cells against cisplatin-induced injury

Conditioned medium of mesenchymal stem cells (MSCs) is now being used for its cytoprotective effects, especially when the cells are equipped with cytoprotective factors to strengthen them against unfavorable microenvironments. Overexpression of Lcn2 in MSCs mimics in vivo kidney injury. Hence, unraveling how Lcn2-engineered MSCs affect kidney cells has been investigated. Cisplatin treated HK-2 or HEK293 kidney cells were co-cultivated with Lcn2 overexpressing MSCs in upper and lower chambers of transwell plates. Proliferation, apoptosis, and expression of growth factors and cytokines were assessed in the kidney cells. Co-cultivation with the MSCs-Lcn2 not only inhibited cisplatin-induced cytotoxicity in the HK-2 and HEK293 cells, but increased proliferation rate, prevented cisplatin-induced apoptosis, and increased expression of growth factors and the amount of antioxidants in the kidney cells. Thus Lcn2-engineered MSCs can ameliorate and repair injured kidney cells in vitro, which strongly suggests there are beneficial effects of the MSCs-Lcn2 in cell therapy of kidney injury.

Mesenchymal stem cells osteogenic differentiation by ZnO nanoparticles and polyurethane bimodal foam nanocomposites

Summary: Mesenchymal stem cells can promote bone repair, especially when used with nano scaffolds/particles. The cytotoxic concentration of Zinc oxide nanoparticles and Polyurethane was evaluated using the MTT and Acridine Orange assay. Results showed that 1% PU scaffold and ZnO NPs boosted the osteogenic differentiation of adipose tissue-derived MSCs, suggesting their potential as a new bone tissue engineering matrix.

Summary: This study aimed to screen and characterize explosive biodegradable Actinomycetes from water, wastewater, and sludge. Actinomycetes isolates from 80 environmental samples were recovered and identified. The results showed that the isolates can degrade and consume 50-80% of RDX and TNT as their sole carbon and energy source. Seeking and screening untapped ecosystems that possess unexplored Actinomycetes will increase the chances of discovering microorganisms capable of degrading TNT and RDX for bioremediation.

Activated mesenchymal stem cells increase drug susceptibility of methicillin-resistant Staphylococcus aureus and Pseudomonas aeruginosa

Summary: This study investigated the use of antibiotics to enhance the antimicrobial effects of mesenchymal stem cells (MSCs) against methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa. Preconditioning MSCs with antibiotics at optimal concentrations improved their ability to inhibit bacterial growth and enhance bacterial clearance. The findings suggest that preconditioned MSCs could be a potential adjuvant treatment for antibiotic-resistant infections.

Accelerating synergistic effects of preconditioned mesenchymal stem cells with Crocin and dexamethasone in pulmonary epithelial cells injury

Summary: This study aimed to investigate the effects of natural (Crocin) and synthetic (Dexamethasone) antioxidants on the efficacy of mesenchymal stem cells (MSCs). The results showed that Crocin and Dexamethasone significantly increased the survival rate of MSCs and A549 cells treated with MSCs. Additionally, they reduced apoptosis and reactive oxygen species (ROS) production, and had an impact on the cytokine levels in treated A549 cells.

Summary: Despite advancements in the prevention, diagnosis, and treatment of colorectal cancer, refractory tumor cells still pose challenges leading to low survival rates and metastasis. Therefore, finding effective and harmless novel molecules for the treatment of colorectal cancer is crucial. Studies have shown that peptides derived from herbal and bacterial sources have antitumor activity with better tolerability and fewer side effects, making them promising candidates for cancer treatment.
