Human foreskin fibroblast-like stromal cells can differentiate into functional hepatocytic cells

Foreskin fibroblast-like stromal cells (FDSCs) are progenitors isolated from human tissue that can differentiate into diverse cell types. Many types of stem cells can differentiate into hepatocyte-like cells, which could be used for drug testing or in liver regeneration therapy, but whether FDSCs can be converted into functional hepatocytes is unknown. FDSCs show divergent properties when cultured in distinct media, forming spheres in Dulbecco's modified Eagle's medium (DMEM) containing F12, epidermal growth factor (EGF), and basic fibroblast growth factor (b-FGF), but have fibroblast-like morphology when cultured in DMEM-based growth medium. Both cell populations express the typical mesenchymal stem cell markers CD90, CD105, and CD73, but the p75 neurotrophin receptor (p75NTR) was detected only in FDSC spheres. Both types of FDSCs can differentiate into hepatocyte-like cells, which express typical liver markers, including albumin and hepatocyte paraffin 1 (Hep Par1), along with liver-specific biological activities. When plasmids containing the human hepatitis B virus (HBV) genome were transfected transiently into FDSCs, differentiated hepatocyte-like cells secrete large amounts of HBe and HBs antigens. FDSCs could be used for clinical hepatic therapy and/or serve as a model of HBV.

Inhibiting TLR7 Expression in the Retinal Pigment Epithelium Suppresses Experimental Autoimmune Uveitis

Summary: This study investigates the role of Toll-like receptor 7 (TLR7) in the development of experimental autoimmune uveitis (EAU). The results show that TLR7 agonist R848 exacerbates EAU progression and impairs the barrier function of retinal pigment epithelial (RPE) cells. The inhibition of TLR7 using shRNA effectively reduces disease severity. These findings highlight the immunomodulatory role of RPE TLR7 and provide a potential therapeutic strategy for autoimmune uveitis.

Summary: The study shows that manganese magnetism-engineered iron oxide nanoparticles (MnMEIO NPs) conjugated with exendin-4 (Ex4) can act as a contrast agent for directly tracing implanted mouse islet beta-cells using magnetic resonance imaging (MRI). This technology can also be used to track implanted porcine neonatal pancreatic cell clusters (NPCCs) containing ducts, endocrine, and exocrine cells. The MnMEIO-Ex4 NPs are excellent contrast agents for detecting and long-term monitoring implanted NPCCs by MRI.

Measurements of Anti-SARS-CoV-2 Antibody Levels after Vaccination Using a SH-SAW Biosensor

Summary: Vaccination is a necessary measure to combat the COVID-19 pandemic, but monitoring antibody levels prior to vaccination is important to reduce unnecessary vaccinations. A novel SH-SAW biosensor can quickly and accurately quantify antibody levels. The study found that antibody concentrations significantly increased several days to weeks after vaccination, with mRNA vaccines achieving higher antibody concentrations.

Exploration of the external and internal factors that affected learning effectiveness for the students: a questionnaire survey

Summary: This study aimed to explore potential factors affecting learning effectiveness in medical technology majors. The results showed that extracurricular studies and willingness to cooperate were positively associated with learning effectiveness, while weakened motivation due to uncertainty was negatively associated. It is suggested that educators should understand students' uncertainty about the future, provide projects requiring joint cooperation and discussion, and focus on content integration rather than rote memorization.

DNA signaturing derived from the internal transcribed spacer 2 (ITS2): a novel tool for identifying Desmodesmus species (Scenedesmaceae, Chlorophyta)

Summary: This study developed a DNA signaturing tool to identify cryptic species in the freshwater green algal genus Desmodesmus. This tool successfully identified 19 species and 24 strain groups, with a signature sequence that could represent almost 90% of Desmodesmus strains. The DNA signaturing tool showed high efficacy in strain identification and accurate classification of cryptic species within the genus Desmodesmus.

DNA signaturing database construction for Tetradesmus species identification and phylogenetic relationships of Scenedesmus-like green microalgae (Scenedesmaceae, Chlorophyta)

Summary: Species identification of Scenedesmus-like microalgae, including Desmodesmus, Tetradesmus, and Scenedesmus, is difficult due to their morphological and genetic similarity. In this study, we developed a DNA signaturing tool for Desmodesmus and constructed a DNA signaturing database for Tetradesmus. This database contains species-specific nucleotide sequences that can identify Tetradesmus strains based on their ITS2 sequences. Our findings also provide insights into the phylogenetic relationships among Scenedesmus-like species and suggest splitting Desmodesmus into two subgenera. The integration of DNA signaturing into a database allows for efficient identification of Scenedesmus-like species through BLAST.

Application of Shear Horizontal Surface Acoustic Wave (SH-SAW) Immunosensor in Point-of-Care Diagnosis

Summary: Point-of-care testing (POCT) has seen explosive growth in the last two decades, with a focus on minimal sample handling, fast results, and low-cost small-sized devices. Shear horizontal surface acoustic wave (SH-SAW) biosensors have emerged as an effective solution for complete whole blood measurements in less than 3 minutes. This review provides an overview of the commercially successful SH-SAW biosensor system for medical use, highlighting its unique features such as disposable test cartridges, mass-produced bio-coatings, and palm-sized readers.

The Upf1 protein restricts EV-A71 viral replication

Summary: This study investigates the potential roles of Upf1 in EV-A71-infected cells. The results suggest that Upf1 is a host protein unfavorable for EV-A71 replication. Knockdown of Upf1 protein increases viral RNA expression and production of progeny virus, while overexpression of Upf1 protein decreases viral RNA expression and production of progeny virus. Additionally, EV-A71 infection suppresses host NMD activity.

The essential role of the ERK activation in large T antigen of BK polyomavirus regulated cell migration

Summary: BK polyomavirus is associated with uro-thelial carcinoma, and TAg and tAg proteins of Merkel cell polyomavirus can induce cell migration through the ERK signaling pathway. The expression of TAg and tAg also regulates the focal adhesion molecules in renal proximal tubular cells. Moreover, TAg regulates MMP-9 expression and activity through ERK signaling, while MMP-2 is regulated through an ERK-independent pathway.
